Optimizing fermentation conditions for cell wall degrading enzymatic production from Trichoderma atroviride SS003 by response surface method

  • ObjectiveThe fermentation conditions of β-glucanase and chitinase production from T. atroviride were optimized to provide references for the research on its biocontrol mechanism. MethodThree independent variables were selected for single factor test , which were incubation time ( 24 , 48 , 72 , 96 , 120 , 144 , 168 hours ) , culture temperature ( 24 , 26 , 28 , 30 , 32 , 34 ℃) and initial inoculums size ( 3 , 4 , 5 , 6 , 7 , 8 , 9 pieces ) . Based on single factor optimization results , response surface method was used to optimize the above conditions in line with Box-Behnken principle of De-sign-Expert 8.05 b. ResultThe optimum conditions for β-glucanase were obtained which were incubation time of 73.70 hours, culture temperature at 27.82 ℃ and initial inoculums size of 5.19 pieces. For chitinase, the optimum conditions were incubation time of 82.91 hours, culture temperature at 27.70 ℃ and initial inoculums size of 5.03 pieces. Con-clusionDue to its effectiveness in boosting quantity of β-glucanase and chitinase from Trichoderma atroviride SS003 , response surface method should be employed to optimize the fermentation conditions for cell wall degrading enzymatic production.
