Sequencing and analysis of the complete mitochondrial genome of Pycnarmon cribrata(Fabricius,1794)(Crambidae: Spilomelinae)

Abstract
【Objective】The study aimed to determine the mitochondrial genome structure characteristics of Pycnarmon cribrata(Fabricius,1794)and analyze its phylogenetic relationship,providing a molecular basis for the taxonomic status and evolutionary relationship of P. cribrata.【Method】The mitochondrial genome of P. cribrata was sequenced by using Illumina Nova 6000 high-throughput sequencing technique,and its genome sequences were annotated and analyzed. Phylogenetic trees were constructed using both maximum likelihood and Bayesian inference methods,based on the mitochondrial protein-coding genes(PCGs)of 21 insect species from 8 subfamilies of Crambidae and 2 insect species from Noctuoidea outgroups.【Result】The mitochondrial genome of P. cribrata was closed double-stranded circular structure,with a total length of 15125 bp. It encoded 37 genes(13 PCGs,22 tRNA genes and 2 rRNA genes)and included 1 A+T-rich region,with 14 intergenic regions and 10 gene overlapping regions. The AT base content in the complete mitochondrial genome sequence of of P. cribrata was 81.1%,representing an obvious AT bias. Among the 13 PCGs,except that cox1 gene had TTG as the start codon,all others began with ATN as the start codon. Except for cox1 and cox2 genes,which ended with the incomplete stop codon T,the remaining genes ended with TAA. In the secondary structures of tRNA,the trnS1 gene lacked DHU arm and loop,and trnT gene lacked the TφC loop. Mismatches of U-U and G-U bases were also existed. The phylogenetic analysis indicated that all species of Spilomelinae belonged to the same branch,confirming Spilomelinae to be a monophyly. P. cribrata,along with P. lactiferalis and P. pantherata from Pycnarmon,gathered into one subgroup,constituting a sister group relationship.【Conclusion】In this study,the complete mitochondrial genome sequence of P. cribrata is obtained. The mitochondrial genome of P. cribrata conforms to the general characteristics of Crambidae mitochondrial genome.
