Transcriptome analysis and juvenile hormone and ecdysone related genes of Citripestis eutraphera at different developmental stages

  • 【Objective】To explore the transcriptome differences of Citripestis eutraphera at different developmental stages, and to explore the growth and development related genes such as juvenile hormone(JH) and ecdysone(Ecd), so as to lay a foundation for the development of the specific prevention and control of C. eutraphera and the development of environmentally friendly growth regulation insecticides. 【Method】High-throughput sequencing technology was used to perform transcriptome sequencing, data assembly, functional annotation and comparative analysis on the larvae, pupae and adults of C. eutraphera. The genes related to JH and Ecd synthesis were screened and verified by real-time fluorescence quantitative PCR. 【Result】The results showed that a total of 254154 unigenes were obtained in the three stages of larvae, pupae and adults. A total of 10688 differentially expressed genes(DEGs) were identified by comparing the larval and pupal stages, and 10041 DEGs were screened by comparing the larval and adult stages. A total of 12896 DEGs were screened out from pupae and adults. The DEGs were compared with Nr database, and the results showed that the sequence similarity with the Amyelois transitella was the highest. GO functional annotation analysis was performed, and a total of 9786 Unigenes were annotated. The DEGs of C. eutraphera at different developmental stages were mostly enriched in cellular structure, cellular process, catalytic activity and binding. There were a total of 14731 unigenes being annotated in the KEGG database, involving 147 metabolism pathways, and most of them were enriched in metabolic pathways and ribosomes, oxidative phosphorylation processes. A total of 49 unigenes related to JH and Ecd synthesis were screened, of which DIB, JHAMT, FPPP, ALDH-1 and ALDH-2 DEGs were analyzed by real-time fluorescence quantitative analysis. ALDH-1,ALDH-2 and DIB expressed highly pupae stage,and the relative expression waw significantly higher than other stages(P<0.05,the same below),FPPP and JHAMT expression was low at larvae stage,and the relative expression was significantly lower than other stages. 【Conclusion】Based on transcriptome analysis, the DEGs of C. eutraphera at various developmental stages are mainly enrich in cellular structure, cellular process, and catalytic activity, with a predominant enrichment to metabolism-related pathways. A total of 49 genes related to JH and Ecd synthesis are screened out. The expression of DIB, JHAMT, FPPP, ALDH-1 and ALDH-2 genes displays significant variations across different developmental stages of C. eutraphera and potentially exert crucial roles in the growth and development of C. eutraphera.
