Composition,diversity and function prediction of intestinal bacteria of Tuta absoluta(Meyrick)larvae in potato population

  • 【Objective】To study composition,diversity and function prediction of intestinal bacteria of Tuta absoluta (Meyrick)larvae in potato population,and to provide a theoretical basis for study on host adaptation mechanism and comprehensive prevention and control of T. absoluta.【Method】Culturable bacteria in T. absoluta intestine were isolated and identified by traditional isolation and culture method and 16S rDNA sequence analysis;full length of 16S rDNA sequences of intestinal bacteria were analyzed by single molecule real-time sequencing technique,and composition structure and diversity of intestinal bacteria were analyzed,as well as function prediction.【Result】There were 5 phyla,13 families,13 genera and 15 species of bacteria in T. absoluta larvae intestine of potato population,and the dominant phylum,family, genus and species were Firmicutes,Enterococcaceae,Enterococcus and E. mundtii. There were 2 phyla,5 families,5 genera and 6 species of culturable bacteria were isolated through traditional isolation. The dominant phylum,family,genus and species were Firmicutes(90.38%),Enterococcaceae(83.71%),Enterococcus(83.71%)and E. mundtii(83.71%). Enterobacter mori(6.56%),Staphylococcus sciuri(5.98%)and Serratia marcescens(2.30%)were the main bacterial species. A total of 5 phyla,6 classes,9 orders,10 families,10 genera and 10 species were annotated by single molecule real-time sequencing technique,and E. mundtii(70.71%)was the dominant species,and E. cloacae(22.16%)was the main bacterial species. ACE,Shannon,Simpson and Chao1 indexes were 7.22±1.91,0.69±0.43,0.28±0.19 and 6.33± 0.94,respectively. Based on the KEGG metabolic pathway analysis,the bacteria from T. absoluta intestine were mainly involved in metabolism pathways of carbohydrate,amino acid,nucleotide and energy. The dominant species E. mundtii was the most important in carbohydrate metabolism and membrane transport.【Conclusion】Bacteria in the intestine of T. absoluta larvae in potato population are abundant,and the dominant species is E. mundtii. These bacteria may participate in carbohydrate,amino acid and nucleotide metabolism.
