Metabolome analysis of Spodoptera frugiperda(J. E. Smith) larvae under chlorantraniliprole stress

  • 【Objective】To understand the response to chlorantraniliprole in Spodoptera frugiperda(J. E. Smith), which could further study the metabolic detoxification of S. frugiperda under chlorantraniliprole.【Method】The metabolome of the 3rd instar larvae of S. frugiperda under the treatment of 0 and 0.10 mg/L chlorantraniliprole exposure were detected by liquid chromatography quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS). The annotation, screening,classification and differentially metabolic pathway analysis of differential metabolites were completed by Progenesis QI softwares and online METLIN databases. The contents of total sugar and soluble sugar in S. frugiperda were determined by microplate reader,and the metabolome results were verified.【Result】A total of 1594 differential metabolites were screened from the metabolome data by positive ion mass spectrometry under 0 and 0.10 mg/L chlorantraniliprole stress,including 853 up-regulated and 741 down-regulated metabolites. A total of 2202 differential metabolites(1102 upregulated and 1100 down-regulated)were identified by negative ion mass spectrometry. The classification of differential metabolites identification results showed that carboxylic acids and derivatives had the largest distribution and content of metabolites and followed by glycerophospholipids. In the identified differential metabolites,levodopa(L-DOPA),phosphatidic acid,succinic acid,serine,maltose,and deoxyuridine were identified,L-DOPA,maltose and deoxyuridine were significantly up-regulated with the difference times of 515.56,7.31 and 9.13(P<0.05,the same below),respectively. Changes were observed in several metabolic pathways including those for amino acids,glycogen,glycolysis,energy,purine,pyrimidine and lipids. Compared with control group,the contents of total sugar and soluble sugar in S. frugiperda treatment group were 2.05 and 1.71 times of those in control group respectively. The results were consistent with the metabolome analysis. The results of KEGG pathway enrichment of the differential metabolites analysis showed that minerals absorption and choline metabolism in cancer pathway were significantly enriched by positive and negative ion mass spectrometry.【Conclusion】Under chlorantraniliprole stress,a large number of differential metabolites of S. frugiperda are greatly changed. The metabolism of amino acid,energy,purine-pyrimidine and fatty acid in the body of S. frugiperda is greatly activated by chlorantraniliprole. These processes together reduce the toxicity of chlorantraniliprole to S. frugiperda.
