Effects of four Chinese herbal medicine compound on antio-xidation and anti-stress indexes of Acipenser dabryanus juveniles

  • 【Objective】To test the effects of four different Chinese herbal medicine compound on the anti-oxidation and anti-stress physiological indexes of Acipenser dabryanus juveniles,so as to provide reference for the promotion and application of traditional Chinese medicine in the breeding and production of A. dabryanus juveniles.【Method】Four medicin componnds were formulated from 17 sources used in traditional Chinese herbal medicines according to the theory of Chinese medicine compatibility. The formulations were:compound I(Houttuynia cordata Thunb:Lonicera japonica Thunb:Rheum palmatum L.:Poria:Glycyrrhiza uralensis Fisch:Astragalus mongholicus Bunge=3:3:1:2:1:2);compound II (Isatis tinctoria:R. palmatum L.:Acanthopanar gracilistμlus W. W. Smith:Codonopsis pilosula=3:3:4:3);compound III(L. japonica Thunb:Prunus armeniaca L.:Cyrtomium fortunei J. Sm:Clerodendrum cyrtophyllum Turcz:Radix sophorae Tonkinensis:Platycodon grandiflorus=3:2:2:2:3:2) and compound IV (A. mongholicus Bunge:Phellodendron:G. uralensis Fisch:Crataegus pinnatifida Bunge:Schisandra chinensis:R. palmatum L.:C. pilosula=3:3:1:2:2:2:2).The basic feed without any Chinese herbal medicine was used as the control group. The compound Chinese herbal medicines were fed at a dosage of 0.5 g/kg body weight during 14 days of the breeding period. Different physiological indexes of juvenile serum were detected on the 7th and 14th days.【Result】Compared with the control group,compounds I and II enhanced the activities of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px),peroxidase (POD),alkaline phosphatase(AKP),the total antioxidant capacity(T-AOC) and content of heat shock protein 70 (HSP70). The contents of malondialdehyde(MDA) and cortisal(COR) of the juvenile serum was reduced to varying degree,but only some differences were significant(P<0.05,the same below). Compounds III and IV clearly enhanced the activities of SOD,CAT,GSH-Px,POD and AKP,the contents of T-AOC and reduced the contents of MDA and COR. Most altered indexes were significant compared with the control. The results of correlation analysis showed that, the remarkably significant posi-tive correlation was found between SOD and GSH-Px,SOD and T-AOC,POD and GSH-Px(P <0.01,the same below). A significant positive correlation was also found between POD and SOD,CAT and T-AOC. Furthermore,both SOD and T-AOC showed a remarkably significant negative correlation with COR. Further significant and negative correlations were observed between GSH-Px and COR,T-AOC and MDA.【Conclusion】Adding suitable Chinese herbal medicine compounds into the basic diet can significantly promote the antioxidant and anti-stress capabilities in A. dabryanus juveniles. In particular, compound III(L. japonica Thunb,P. armeniaca L.,C. fortunei J.Sm,C. cyrtophyllum Turcz,R. sophorae Tonkinensis) and compound IV(A. mongholicus Bunge,Phellodendron,G. uralensis Fisch,C. pinnatifida Bunge,S. chinensis,R. palmatum L.,C. pilosula) have the best effect. These formulations can effectively relieve and protect A. dabryanus juveniles against stress.
