SSR analysis for genetic diversity of Angelica dahurica var. formosana

  • ObjectiveThe genetic diversity of Angelica dahurica var.formosana gerplasm resources was analyzed to provide reference for sustainable utilization and new varieties breeding of A.dahurica gerplasm resources.MethodSix pairs of polymorphic SSR primers were screened from the published 18 pairs of SSR primers in A.sinensis,another An-gelica species.Fifty-three samples from three populationsKZ(Kunming),NZ(Nanjing),XY(Xianyang)of A.dahurica were amplified by PCR.The genetic diversity,genetic difference and cluster analysis were conducted by GENEPOP 4.0, GenALeX 6.4.1 and Structure 2.2 respectively based on their genotype data.ResultIn all six SSR loci,a total of 37 al-leles were amplified,the average allele number(Na)was 6.2,effective allele number(Ne)1.798,and Shannon informa-tion index(I)1.802.The observed heterozygosity(Ho),expected heterozygosity(He)and gene flow(Nm)were 0.245, 0.304 and 1.340,respectively.Furthermore,the genetic diversity among the three populations presented some differences. KZ population had the highest genetic diversity(Na 3.8,Ho 0.481 and He 0.508).The three populations were clustered in-to two groups:KZ population was in one group and NZ and XY populations were in another.It indicated that genetic rela-tionship between XY population and NZ population was close.The genetic difference analysis presented that,genetic dif-ferentiation index(FST)between XY population and NZ population was low(0.208),which indicated that genetic differen-tiation level between the two was low and genetic difference was small.FST between KZ and NZ populations(0.510),KZ and XY populations(0.506)were high,which meant the genetic differentiation levels for them were high and genetic di-fference was great.ConclusionSSR primers of different species belong to the same genus may be universalizable. A.da-hurica populations from different places present certain genetic differentiation,but genetic diversity in general is low. Therefore,it is necessary to search closely related wild resources of A.dahurica to ensure the sustainable utilization of an-gelica root resources.
