Isolation and identification of Jinhua fungus in Liupao tea

  • ObjectiveJinhua fungus in Liupao tea was identified in order to provide theoretical basis for Liupao tea in-dustry development. MethodSpread-plate method and three-point inoculation method were adopted to isolate and purify Jinhua fungus from Liupao tea produced by China Tea(Wuzhou) Factory in November, 2015. The colony characteristics were observed by traditional morphology observation method, and the structures like hypha and conidium were observed via optical microscope. Then DNA sequence analysis for strain isolated was conducted combining molecular biology method, and homology searching and comparison via ITS sequence and phylogenetic tree analysis were carried out. ResultOne strain of Jinhua fungus was isolated and named LB-1. The morphological characteristics of LB-1were similar to those of Eurotium:golden colony, concentric rings, radioactive ridge, etc. The homology of its ITS sequence to Eurotium amstelodamiwas 99%. Therefore , LB-1 was identified as E. amstelodami. ConclusionJinhua fungus LB-1 is isolated from Liupao tea , and identified as E. amstelodami in Eurotium. During processing of Liupao tea, the producers can create suitable environ-ment for LB-1 and promote the mass reproduction of LB-1, so as to enhance the health care effects of Liupao tea.
