Abstract
ObjectiveThe experiments were conducted to find out the suitable culture medium for the seeds germination,bud multiplication,roofing and growth of plantlet in Dendrobium candidum Wall.ex Lindl,and to develop a technique for its rapid propagation. MethodSeeds of Dendrobium candidum were used as explants in tissue culture,and the effects of different media with different concentrations of 6-BA (0-3.0 mg/L) and NAA (0.1-0.3 mg/L),supplemented with mashed banana (15-25%) or mashed potato( 15-25% ),on the seed germination,bud multiplication,rooting and growth of plantlets were investigated. ResultThe most suitable culture medium for seed germination and bud multiplication was MS+20% mashed potato and MS+6-BA (2.0 mg/L),respectively. However,the best suitable medium for rooting was found MS+15% mashed banana which gave 100% rooting rate. ConclusionThe strong rooting plantlet can be obtained in medium supplemented with mashed potato and mashed banana. This medium may be recommended for the large scale production of Dendrobium candidum plantlets.
