Germination and induction of axillary buds in micropropagated Lonicerae hypoglauca

Abstract
The shoot of Lonicerae hypoglauca cuhivar (Lonicerae confusa DC) with axillary buds was used as explant to study the effects of different stefihzation methods and sampling seasons, and different concentrations of hormones and active ingredient on the germination, induction and proliferation of axillary buds in micropropagated shoots. The sterilization treatment with 75% alcohol for 25 s combined with dipping shoots in 0.1% HgCl2 solution with tween-80 for 6 rain wasfound to be the most effective, as shown by 53.3% germination rate of axillary buds. Further, the sterilization effects were found to be most effective in spring sampled explants, which germinated after 7 days of culture, compared to 12 days in autumn samples. The suitable culture medium for germination and proliferation of axillary buds were found as follows: MS media+6-BA (3.0 mg/L) + KT(0.5 rag/L), and MS media+6-BA (2.0 mg/L) + KT(0.5 mg/L), respectively.
