宫粉羊蹄甲花粉离体萌发培养基的筛选

Screening of culture medium for in vitro pollen germination of Bauhinia variegate L.

  • 摘要: 目的筛选最适宫粉羊蹄甲花粉离体萌发的培养基及培养条件,为其花粉活力检测、品种繁育及苗木生产提供参考依据.方法宫粉羊蹄甲花粉粒采用液态培养法,设置培养基组分、pH和温度等8个影响因子的梯度值进行逐步迭代优化,以缺素培养为对照(CK),探讨其花粉萌发效果.结果培养基各组分对宫粉羊蹄甲花粉离体萌发的影响效果迥异,其中H3BO3对花粉萌发起决定性作用,花粉萌发对蔗糖较敏感.宫粉羊蹄甲花粉在添加8.0%蔗糖、10.0%聚乙二醇-6000(PEG-6000)、0.0020% H3BO3、0.02% CaC12·2H2O、0.02% MgSO4· 7H2O及0.010%KNO3的液体培养基中,于pH 6.8、恒温29℃条件下培养约20 min可观察到最佳萌发效果,萌发率均值达91.230%.结论宫粉羊蹄甲花粉在液体培养基各组分及培养条件最佳组合中离体培养才能提高萌发率,花粉管才能正常生长.
