抗金银花白粉病菌贝莱斯芽孢杆菌HC-8菌株培养基及发酵条件优化

Optimization of culture medium and fermentation parameters of Bacillus velezensis HC-8 antagonistic to Erysiphe lonicerae

  • 摘要: 【目的】优化抗金银花白粉病菌贝莱斯芽孢杆菌HC-8菌株的发酵培养基及发酵条件,为其快速大量生产及进行大田生物防治提供理论依据。【方法】以贝莱斯芽孢杆菌HC-8菌株为材料,采用单因素试验和正交试验,分别优化其发酵培养基及发酵条件;结合分生孢子萌发和平板对峙试验,分析优化方案下HC-8菌株对白粉病菌分生孢子萌发的抑制效果及对尖孢镰刀菌、烟草疫霉和轮状镰刀菌的拮抗活性。【结果】HC-8菌株最适基础培养基为酵母蛋白胨培养基(YSP);最佳培养基配方为蛋白胨5.0 g/L、酵母膏25.0 g/L、麦芽糖10.0 g/L;最佳发酵条件为温度37℃、初始pH 6.0、转速180 r/min、接种量0.100%、装液量20 mL/300 mL、培养时间48 h。优化方案下,HC-8菌株对白粉病菌分生孢子萌发的抑制率为83.15%,显著高于优化前的74.65%(P<0.05,下同);对尖孢镰刀菌、烟草疫霉和轮状镰刀菌的抑菌率分别为60.66%、59.03%和65.32%,显著高于优化前的54.31%、55.24%和59.17%。【结论】优化后的方案可提高HC-8菌株的发酵产量,并增强对尖孢镰刀菌、烟草疫霉和轮状镰刀菌的拮抗效果及对白粉病菌分生孢子萌发的抑制效果。建立的方案可用于快速、大批量发酵HC-8菌悬液。

     

    Abstract: 【Objective】 To provide a theoretical basis for rapid mass production and executing field biocontrol of Bacillus velezensis HC-8 strain antagonistic to Erysiphe lonicerae, it is necessary to optimize its fermentation media and conditions.【Method】 In this study, the strain HC-8 was used as the material, and the single factor experiment and orthogonal test were employed to optimize the fermentation media and conditions; combined with conidial germination assays and plate confrontation test to analyze the inhibition effect of strain HC-8 on conidial germination of E. lonicerae and antifungal activity of strain HC-8 against Fusarium oxysporum, Phytophthora nicotianae and F. verticillioides under the optimized fermentation conditions.【Result】 The most suitable media for HC-8 strain was yeast saccharose peptone media (YSP); the best media formula was peptone concentration of 5.0 g/L, yeast extract concentration of 25.0 g/L and maltose concentration of 10.0 g/L; and the most suitable culture conditions were the fermentation temperature of 37 ℃, the initial pH of 6.0, the shaking speed of 180 r/min, the inoculation volume of 0.100% (v/v), the liquid medium volume of 20 mL/ 300 mL and the incubation time of 48 h. Moreover, strain HC-8 showed inhibition rates of 83.15% on E. lonicerae conidial germination under the optimized fermentation conditions, significantly higher than of 74.65% before optimization(P<0.05, the same below). Furthermore, strain HC-8 displayed inhibition rates of 60.66%, 59.03% and 65.32% against F. oxysporum, P. nicotianae and F. verticillioides, respectively, under the optimized fermentation conditions, which were significantly higher than of 54.31%, 55.24% and 59.17%, respectively, under the condition of before optimization. 【Conclusion】 The optimized medium and fermentation conditions can effectively increase the fermentation yield and antagonistic activity of strain HC-8 against F. oxysporum, P. nicotianae and F. verticillioides, as well as the inhibition on E. lonicerae conidial germination. The optimized scheme can be used in rapid and mass production of fermented HC-8 bacterial suspension.
