一株香蕉枯萎病拮抗菌的筛选、鉴定及生防效果研究

Screening,identification and biocontrol effect of antagonistic bacteria against banana Fusarium wilt

  • 摘要: 【目的】筛选获得对香蕉枯萎病病原菌具有拮抗作用的菌株,为香蕉枯萎病生物防治提供菌株资源。【方法】以香蕉根际土壤为材料,利用铬天青(Chromeazurol S,CAS)双层平板法和平板对峙法筛选可分泌铁载体且对香蕉枯萎病菌具有拮抗作用的菌株;通过平板对峙法测定拮抗菌株对6种常见植物病原菌的拮抗活性;通过形态学观察、生理生化测定和16S rDNA序列分析明确拮抗菌株的分类地位;通过盆栽试验验证拮抗菌株对香蕉枯萎病的防治效果及对香蕉植株的促生作用。【结果】经CAS培养基初筛,从土壤中分离获得60株可分泌铁载体的菌株,平板对峙法复筛得到5株拮抗作用较强的菌株,其中菌株Gxun-2的抑菌效果最强,平均抑菌率达74.45%,且对供试的其他6种植物病原菌均具有较好的抑制效果,具有广谱抗菌性。结合菌落形态、生理生化及16S rDNA进化树分析结果,菌株Gxun-2鉴定为铜绿假单胞菌(Pseudomonas aeruginosa)。盆栽试验显示,菌株Gxun-2对香蕉枯萎病具有较好的防病作用,防治效果达84.21%,且对香蕉植株具有一定的促生效果,香蕉苗鲜重较对照增加32.96%。【结论】菌株Gxun-2对香蕉枯萎病具有较好的防治作用,且对香蕉植株具有一定的促生效果,可作为研制香蕉枯萎病生防菌剂的候选菌株资源。

     

    Abstract: 【Objective】 The aim was to screen and obtain antagonistic strains against banana Fusarium wilt, and provide strain resources for biological control of banana Fusarium wilt.【Method】 The banana rhizosphere soil was used as the material to screen the strains that could secrete siderophores and had antagonistic effect on Fusarium oxysporum f sp.cubense by using chromeazurol S(CAS) double-layer plate and plate confrontation method. The antagonistic activity of antagonistic strains against six common plant pathogenic bacteria was determined by plate confrontation method. The taxonomic status of the antagonistic strains was clarified through physiological and biochemical determination and 16 S rDNA sequence analysis. The pot experiment was conducted to verify the efficacy of the antagonistic strains against banana Fusarium wilt and their growth-promoting effects on banana plants.【Result】 After initially screened with CAS medium, 60 strains secreting siderophores were screened out from the soil, and five strains were selected out of them by plate confrontation method. Among them, Gxun-2 had the highest bacteriostatic rate, reaching 74.45%, and had fine inhibitory effect on the other six plant pathogens tested, with broad-spectrum antibacterial property. Combined with colony morphology, physiology and biochemistry and 16 S rDNA phylogenetic tree analysis, Gxun-2 was identified as Pseudomonas aeruginosa. Pot experiment showed that the strain had obvious disease prevention effect on banana Fusarium wilt with the control effect reaching 84.21%. It had certain growth promoting effect on banana plants, the fresh seedling weight increased by32.96%.【Conclusion】 Gxun-2 strain has a strong effect on preventing and treating banana wilt, and promoting growth of banana seedlings, and can be used as a candidate strain resource for development of candidate strain resources for biological control of banana Fusarium wilt.
