秀珍菇退化菌株生物学特征比较及dsRNA病毒检测

Biological characteristics of degenerated strains of Pleurotus pulmonarius and detection of dsRNA virus

  • 摘要: 目的比较秀珍菇退化菌株与正常菌株的生物学特征并进行双链RNA(dsRNA)病毒检测,为秀珍菇栽培过程中选择优质菌种提供参考依据.方法开展秀珍菇退化菌株菌丝拮抗、菌丝生长速度及出菇试验,与正常菌株进行生物学特征比较,通过ITS扩增,利用RAPD、ISSR和SRAP分子标记分别进行差异性比较,并开展dsRNA病毒检测,分析秀珍菇菌株的退化原因.结果秀珍菇退化菌株X9和X13与正常菌株X5、X6及对照菌株X15间存在拮抗反应,当培养基的pH低于5.0时,退化菌株的菌丝生长速度显著降低(P<0.05,下同).退化菌株X13菌丝的生长速度(4.95mm/d)、常温出菇单袋产量(73.34g/袋)及低温刺激出菇单袋产量(107.69g/袋)均显著低于正常菌株和对照菌株.分子标记分析未发现供试菌株间存在遗传差异,dsRNA病毒检测发现供试菌株均存在dsRNA片段,其中退化菌株X13存在特异的dsRNA片段.结论秀珍菇退化菌株可能受dsRNA病毒感染,适应环境能力变弱,产量降低,生产上应注意菌株来源,避免使用退化菌株.

     

    Abstract: ObjectiveBiological characteristics of degenerated strains and normal strains of Pleurotus pulmonarius were compared and double-stranded RNA(dsRNA)virus detection was conducted to offer references for choosing pre-mium strains in cultivations of P.pulmonarius.MethodAntagonism reaction test,mycelium growth rate and fruiting ex-periment were carried out on degenerated strains,and its biological characteristics were compared to the normal stains.By amplification of ITS,difference comparison were carried out using RAPD,ISSR and SRAP molecular marker.The rea-sons of degradation of P.pulmonarius were studied by dsRNA virus detection.ResultAntagonism reaction was found be-tween degenerated strains(X9 and X13)and normal strains(X5,X6 and X15).When pH of culture medium was lower than 5,the mycelium growth rate of degenerated strains significantly(P<0.05,the same below)decreased.The mycelium growth rate(4.95 mm/d),normal temperature fruiting yield per bag(73.34 g/bag)and low temperature stimulate fruiting yield per bag(107.69 g/bag)of degenerated strains X13 were significantly lower than those of normal strains and control strains.According to molecular marker analysis,there was no genetic difference among the tested strains.dsRNA frag-ments were found in all tested strains through dsRNA virus detection.Specific dsRNA fragments were detected in degene-rated stain X13.ConclusionThe degenerated strains of P.pulmonarius may be affected by dsRNA virus,resulting in weak ability to adapt to the environment and low yield.Strain source should be attached great importance to and degenera-ted stains should be avoided during cultivation.
