稻曲病菌薄壁分生孢子产孢平板培养基的筛选

Screening plate media for sporulation of thin-walled conidia of Ustilaginoidea virens(Cooke)Takahashi

  • 摘要: 目的分析平板培养基的组分构成对稻曲病菌薄壁分生孢子形成的影响,为薄壁分生孢子的制备培养提供便捷的技术手段.方法以4个常规稻曲病菌株Uv-34、Uv-105、Uv-110和Uv-111为试验材料,采取常规的固体培养技术程序,分别测试马铃薯蔗糖琼脂培养基(PSA)的马铃薯含量及蔗糖含量对薄壁分生孢子形成的影响、在水(非糖)和4种糖(蔗糖、核糖、木糖和山梨糖)与马铃薯组配形成的固体培养基(PA、PSA、PRA、PXA和PSoA)上的产孢量、在固体培养基上形成孢子数量的时间变化动态及在PA固体培养基上4个菌株的产孢表现.结果在PSA培养基中,蔗糖含量为10g/L时产孢量最高(1.69×106个/mL),马铃薯含量以50.0g/L时的产孢量最高(1.18×106个/mL).在PA、PSA、PRA、PXA和PSoA等平板培养基上,菌株的产孢量分别为6.98×106、4.63×106、11.92×106、0.84×106和0.01×106个/mL.在15d的连续培养中,于PSA培养基上培养5d后产孢量开始大幅下降,而在PA和PRA培养基上菌株产孢量基本稳定或保持上升趋势.在PA培养基上,4个供试稻曲病菌株均能正常大量产孢.结论稻曲病菌能在含马铃薯的常规固体培养基上产孢;PA是日常制备培养稻曲病菌薄壁分生孢子的简单易用培养基;PRA是制备培养薄壁分生孢子的高产培养基.

     

    Abstract: ObjectiveEffects of component and content of plate medium on sporulation of thin-walled conidia of Ustilaginoidea virens(Cooke)Takahashi was examined in order to provide an easy and rapid technical way for production of thin-walled conidia.MethodFour conventional U.virens strains(Uv-34,Uv-105,Uv-110 and Uv-111)were used as materials.Using common plate culture technical procedure,effects of both potato content and sucrose content in medium potato sucrose agar(PSA)on sporulation of thin-walled conidia was determined,conidia yield of strains grown on plate medium(PA,PSA,PRA,PXA and PSoA)of potato mixed with water(no sugar)and four sugars(sucrose,ribos,xylose and sorbose)was observed,variation trend of conidia yield produced on plate medium due to the change of culture time was recorded,and sporulation of four strains on plate medium of potato agar(PA)was observed.ResultOn PSA medi-um,the top yield(1.69×106conidia/mL)of conidia was obtained from 10 g/L of sucrose,and top yield(1.18×106conidia/mL)of conidia was obtained from 50 g/L of potato.On plate media PA,PSA,PRA,PXA and PSoA,conidia yields were 6.98×106,4.63×106,11.92×106,0.84×106 and 0.01×106conidia/mL.Conidia yield produced on PSA plate media dropped sharply 5 d after culture,but conidia yield produced on both PA plate media and PRA plate media remained steady or increasing trend within a culture duration of 15 d.All four strains produced conidia normally and abundantly on PA plate media.ConclusionU.virens can produce conidia on conventional plate media with potato on.PA is a medium which is quick and easy to use in daily work for the production of thin-walled conidia of U.virens.PRA is a medium to produce thin-walled conidia with high yield.
