烟草种质资源TI203对烟草花叶病的抗性特点

Resistance characteristics of tobacco germplasm resource TI203 to Tobacco mosaic virus

  • 摘要: 目的系统了解烟草种质资源TI203抗烟草花叶病毒(Tobacco mosaic virus,TMV)的特点,为烟草抗病育种的亲本选择提供更丰富信息.方法以烟草种质资源TI203为材料,采用TMV病毒接种、病情调查、病毒外壳蛋白累计量检测、TMV复制水平测定及日灼现象观察等方法,研究TI203对TMV的抗性特异性、TI203接种TMV普通株系(TMV-U1株系)后的症状、病毒复制水平等.结果TI203只对TMV表现无症状的抗病特点,接种马铃薯Y病毒(PVY)和黄瓜花叶病毒(CMV)则表现出典型症状.接种TMV-U1株系后14 d,Western blotting检测结果显示,TI203叶片病毒复制水平明显低于对照品种;用表达GFP的TMV侵染性克隆浸润接种发现,TI203对TMV的抗性发生在病毒复制水平.在高温和强日照条件下,TI203生长迅速的中上部叶片会发生日灼现象,且难以恢复.结论TI203只对TMV表现无症状,其对TMV的抗性发生在病毒复制水平.

     

    Abstract: ObjectiveThis research was conducted to understand the resistance characteristics of tobacco germplasm resource TI203 to Tobacco mosaic virus(TMV),in order to provide more information for parent selection in disease resis-tant breeding of tobacco.MethodTobacco germplasm resource TI203 was taken as experimental material. The resistance specificity of TI203 to TMV,and symptom and level of virus replication of TI203 after inoculated with TMV ordinary strain(TMV-U1 strain)were studied by TMV inoculation,disease investigation,virus coat protein accumulation detec-tion,TMV replication level determination and observation on sunburn phenomenon.ResultThe results showed that TI203 exhibited a symptomless reaction to TMV,but typical symptoms to Potato virus Y(PVY)and Cucumber mosic vi-rus(CMV)after inoculation. Western blotting detection results indicated that the level of virus replication in TI203 leaves was obviously lower than that of control variety 14 d after inoculated with TMV-U1 strain. Resistance of TI203 to TMV was observed at virus replication level by using TMV-GFP infectious clone infiltration inoculation. Under high tempera-ture and strong sunlight conditions,sunburn phenomena always occurred on fast-growing middle and upper leaves of TI203,and the leaves were difficult to recover.ConclusionTI203 is symptomless to TMV and its resistance to TMV occurs at virus replication level.
