辣椒疫病罹病植株根际土壤细菌群落多样性分析

Comparison on bacterial community diversity in rhizosphere soil of peppers with phytophthora blight

  • 摘要: 目的分析辣椒疫病罹病植株根际土壤细菌群落多样性,为辣椒疫病防控提供理论依据.方法采集湖南长沙、吉首及永州地区辣椒疫病罹病植株根际土壤,运用IlluminaMiSeq高通量测序技术对土壤样品基因组DNA的16S rDNA-V4区进行测序分析,明确不同地区罹病辣椒植株根际土壤细菌群落多样性.结果3个地区罹病辣椒根际土壤的细菌OTU数量存在差异,独有OTU占总体比例各有差异,其中永州地区占其总数的30.18%、吉首地区占其总数的41.83%、长沙地区占其总数的24.07%,3个地区共有OTU占所有检测到OTU的14.95%.α多样性指标均表现为长沙地区最低,吉首地区最高.在门水平上,3个地区的罹病辣椒根际土壤微生物组成相似,优势菌门为Acidobacteria(酸杆菌门)、Proteobacteria(变形菌门)、Actinobacteria(放线菌门)和Chloroflexi(绿弯菌门),但3个地区在相对丰度上表现一定差异.结论湖南永州、吉首和长沙地区辣椒疫病罹病植株根际土壤细菌多样性存在差异,以吉首最高、长沙最低;3个地区的罹病辣椒根际土壤微生物组成相似,但相对丰度存在差异.

     

    Abstract: ObjectiveIn the present study, bacterial community diversity in rhizosphere soil of peppers with phytoph-thora blight was studied to provide reference for control of pepper phytophthora blight. MethodRhizosphere soil of peppers dying from phytophthora blight was collected from Changsha, Jishou and Yongzhou in Hunan. 16S rDNA-V4 regions of sample genome DNA were sequenced by IlluminaMiSeq high-throughput sequencing to analyze bacterial community diver-sity in rhizosphere soil of peppers with phytophthora blight in different areas. ResultBacteria OTU number and propor-tions of unique OTU of rhizosphere soil of peppers with phytophthora blight of three areas were different. The proportion of Yongzhou accounted for 30.18% of the total, and that of Jishou accounted for 41.83% of the total and that of Changsha accounted for 24.07% of the total. Shared OTU of the three areas accounted for 14.95% of all detected OTU. Alpha diversity index were the lowest in Changsha and the highest in Jishou. At phylum level, the microbial composition was similar in rhi-zosphere soil of peppers with phytophthora blight of three areas, the dominant bacteria were Acidobacteria, Proteobacteria, Actinobacteria and Chloroflexi, but relative abundance showed some differences. ConclusionThere is difference in bac-terial community diversity in rhizosphere soil of peppers dying from phytophthora blight in Yongzhou, Jishou and Chang-sha, the diversity of Jishou is the highest while that of Changsha is the lowest. The microbial composition is similar in rhizo-sphere soil of peppers with phytophthora blight of the three areas, but the relative abundance is different.
