冬种马铃薯试管苗壮苗技术研究

Test tube cultivation technology for culturing strong potato plantlets suitable for winter planting

  • 摘要: 目的建立冬种马铃薯试管苗高效生产技术体系.方法在不同植物生长调节剂、光强和昼夜温差、培养器皿和封口材料、固液体MS和1.5MS培养基条件下对马铃薯试管苗进行壮苗培养.结果在MS培养基中添加B9和CCC,均对马铃薯试管苗壮苗培养有促进作用,但以B9的效果最佳、最持久;当光照强度为3000 lx、昼夜温差为8℃左右时,即培养温度在18~26C时壮苗培养效果较佳.与固体培养基相比,1.5MS液体培养基对试管苗的壮苗效果更明显;与试管和250 mL罐头瓶相比,透光、透气性好的150 mL三角瓶带塑料封口膜更有利于马铃薯试管苗壮苗培养,且操作方便,试管苗生长较好.结论在光照强度3000 lx、温差为8℃条件下,在带塑料封口膜的150 mL三角瓶中采用添加B9的1.5液体MS培养基对马铃薯试管苗进行壮苗培养,可获得较好的培养效果.
