Abstract:
ObjectiveIn order to provide scientific reference for extraction and application of polyphenols from Sar-gassum fusiforme, the enzyme-assisted extraction process of polyphenols from S. fusiforme was optimized by orthogonal ex-perimental method to improve yield of polyphenols. MethodWith fresh S. fusiforme as raw material, the polyphenols were extracted from S. fusiforme by enzyme-assisted extraction method. The effects of compound enzyme mass ratio(neutral pro-tease amount: cellulase amount), enzymolysis temperature, enzymolysis pH and enzymolysis time on extraction yield of polyphenols were studied through single factor experiment, and then extraction process conditions were optimized by or-thogonal experimental method. The enzyme-assisted extraction method was compared with traditional solvent extraction method. ResultThe results showed that, the factors influencing extraction yield of polyphenols were in order as follows:enzymolysis pH>enzymolysis temperature>compound enzyme mass ratio>enzymolysis time. And enzymolysis pH and tem-perature had significant effects on extraction yield of polyphenols(P<0.05). The optimum enzymolysis conditions were as follows: enzymolysis temperature of 50 ℃, enzymolysis pH of 5.5, enzymolysis time of 45 min, and compound enzyme mass ratio of 20∶1(126 mg/g composite enzyme). Under above optimum conditions, the extraction yield of polyphenols from S. fusiforme was up to 9.26 mg/g, and obviously higher than that(8.26 mg/g) of solvent extraction method. ConclusionEnzyme-assisted extraction process optimized by orthogonal experiment method can effectively improve extraction yield of polyphenols from S. fusiforme, and the optimized process parameters can be applied in actual production.
