Abstract:
ObjectiveThe in vitro tissue culture technique of oriental lily was studied, and the unified culture medi-um was screened, in order to provide theoretical and technical support for industrialized seedling production. MethodThe bulb scales of three lily cultivars viz., Tiber, Sorbonne and Siberi were used as explants, which were cultured in minimal medium(MS) supplemented with different hormones. ResultThe results showed that, the best sterilization method was as follow: 0.1% HgCl2 for 12 min, followed by 75% alcohol for 15 s. The suitable culture medium for callus induction and differentiation was MS. The optimum medium for subculture multiplication was MS+0.5 mg/L6-BA+0.5 mg/L NAA , and the multiplication coefficients of Tiber, Sorbonne and Siberi were 3.32, 3.32 and 3.00, respectively. The optimum culture medium for rooting culture was MS+0.1 mg/L IBA, and the rooting rates of Tiber, Sorbonne and Siberi were 96.33%, 92.67%, 93.67% respectively. The optimum substrate for transplanting was peat mixed with perlite in the volume propor-tion of 2∶1, the survival rates of Tiber, Sorbonne and Siberi were 97.34%, 98.00% and 100.00%, respectively. Conclu-sionThe tissue culture and rapid propagation system for 3 varieties of oriental lily is established , and which is applicable to factory production of lily bulb.
