Abstract:
【Objective】 The aim of the study was to clarify the resistance level of the introduced indica rice cultivars from abroad against rice blast disease in the rice-growing areas of Yunnan, as well as the composition of known resistance genes, which could provide reference for the selection and breeding of new disease-resistant varieties using resistance resources, as well as for the discovery of new disease-resistant genes.【Method】 Panicle blast resistance of 258 introduced foreign indica rice cultivars was evaluated through 2 consecutive years (2023-2024) of field screening under natural blast nursery, subsequently, the highly resistant indica rice cultivars against panicle blast were inoculated with 17 single-spore strains of rice blast pathogens at seedling stage, and indoor leaf blast resistance was identified. Molecular detection of eight known resistance genes located at four loci (
Piz-t,
Pi2,
Pi9,
Pib,
Pi1,
Pita-2,
Pikp, and
Pikh) was performed using functional molecular markers.【Result】 In the blast nursery, 19, 2, 21, 77, 51, and 88 indica rice cultivars were classified as highly resistant, resistant, moderately resistant, moderately susceptible, susceptible, and highly susceptible respectively among the 258 introduced foreign indica rice cultivars. Nineteen highly-resistant indica rice cultivars were subjected to artificial inoculation at the seedling stage using 17 dominant single-spore strains of rice blast pathogen, and 11 indica rice cultivars of them exhibited resistance to all strains. Among the 258 introduced foreign indica rice cultivars, 125 cultivars carried 1 or multiple target resistance genes, accounting for 48.45% of the tested cultivars. Of these, 93 cultivars carried 1 target gene, 27 cultivars carried 2 target genes, and 5 cultivars carried 3 target genes. Among the 8 target genes analyzed,
Pib,
Pita-2,
Pi2,
Pi1,
Pikp,
Pikh, and
Piz-t genes were detected in 85, 40, 15, 8, 5, 5, and 4 indica rice cultivars respectively, accounting for 32.95%, 15.50%, 5.81%, 3.10%, 1.94%, 1.94%, and 1.55% of all tested cultivars. Among all detected genes, the resistence gene
Pib showed the highest distribution frequency, whereas
Piz-t, Pikp, and
Pikh genes were present at relatively low frequencies, and
Pi9 gene was not detected in any cultivar.【Conclusion】 From 258 introduced foreign indica rice cultivars, 11 cultivars that show broad-spectrum resistance to rice blast disease in Yunnan are selected. These 11 broad-spectrum resistant indica rice cultivars can be used as excellent sources of resistance for disease resistance breeding and the exploration of resistance genes in the rice-growing areas of Yunnan.
