不同西番莲品种(系)的高温响应及耐热性评价

High-temperature response and heat tolerance evaluation of different passion fruit varieties (lines)

  • 摘要: 【目的】 探究不同西番莲品种(系)对高温的响应并开展耐热性综合评价,为西番莲高产稳产及培育耐热性西番莲品种提供理论参考。【方法】 以12个西番莲品种(系)为试验材料,采用双因素裂区试验设计,主区因素为温度,设置高温处理和对照(露天种植);裂区因素为品种(系)。测定叶片的相对电导率、花粉萌发率、可溶性蛋白和可溶性糖含量以及过氧化氢酶(CAT)、超氧化物歧化酶(SOD)和过氧化物酶(POD)活性等7个生理指标,测定单株挂果数和坐果率2个农艺性状指标,并分析9个指标间的相关性。采用隶属函数法对12个西番莲品种(系)的耐热性进行综合评价及聚类分析。【结果】 与对照相比,高温处理40 d后西番莲叶片的SOD活性显著降低(P<0.05,下同),POD活性极显著降低(P<0.01,下同),CAT活性极显著增加;相对电导率、可溶性糖和可溶性蛋白含量均无显著差异(P>0.05)。高温处理40 d后供试西番莲品种(系)的平均花粉萌发率、高温处理0~40 d平均单株挂果数、盛花期平均坐果率分别较对照降低4.55%、45.38%和32.27%。品种(系)间比较结果显示,P5、H3和H36的花粉萌发率为78.88%~82.42%,显著高于其他品种(系),P1、DN和夏纪的花粉萌发率仅为31.33%~40.58%,显著低于其他品种(系);高温处理0~40 d,MT和H36的单株挂果数分别为12.00和7.57个,MT显著高于其他西番莲品种(系),H3和MT盛花期的坐果率分别为66.39%和51.14%,显著高于其他品种(系)。相关分析结果表明,坐果率分别与SOD和POD活性以及花粉萌发率呈显著正相关。根据耐热性的综合评价值(D),12个西番莲品种(系)的耐热性排序为H36>MT>台农1号>H3>P5>P7>夏纪>P1>冬荟蜜>圆小>黄优2号>DN。聚类分析结果显示,12个西番莲品种(系)分为四大类,其中H36和MT的耐热性较强,黄优2号和DN的耐热性较差。【结论】 花粉萌发率、坐果率和单株挂果数可用作西番莲不同品种(系)的耐热性综合评价指标。筛选出2个耐热性较强的西番莲不同品种(系),分别为H36和MT。

     

    Abstract: 【Objective】 This study aimed to investigate the responses of different passion fruit varieties (lines) to high temperature and conduct comprehensive evaluation of their heat tolerance,so as to provide theoretical references for high and stable yield cultivation and breeding of heat-tolerant varieties for passion fruit. 【Method】 Using 12 passion fruit varie-ties (lines) as experimental materials,and a two-factor split-plot experimental design was adopted: the main plot factor was temperature, for which a high-temperature treatment and an open-field control were set; variety (line) was the split-plot factor. Seven physiological indicators of leaf leaf relative conductivity, pollen germination rate, soluble protein content, soluble sugar content, catalase (CAT) activity, superoxide dismutase (SOD) activity, and peroxidase (POD) acti-vity and two agronomic traits (fruit set per plant and fruit setting rate) were determined; and the correlations among the nine indicators were analyzed. The comprehensive evaluation and cluster analysis of heat tolerance of the 12 passion fruit varieties (lines) was performed using membership function method. 【Result】 After 40 d of high-temperature treatment and compared with control, the SOD activity in passion fruit leaves significantly decreased (P<0.05, the same below), the POD activity extremely significantly decreased (P<0.01,the same below), and the CAT activity extremely significantly increased; no significant differences were observed in relative conductivity, soluble sugar content, and soluble protein content between the high-temperature treatment and the control (P>0.05). After 40 d of high-temperature treatment, the average pollen germination rate, average fruit set per plant within 0-40 d of treatment,and average fruit setting rate at full bloom of the tested passion fruit varieties (lines) were 4.55%, 45.38%, and 32.27% lower than those of the control, respectively. According to comparison results among the varieties, P5, H3, and H36 had pollen germination rates of 78.88%-82.42%, which were significantly higher than those of other varieties (lines); while P1, DN, and Xiaji had pollen germination rates of just 31.33%-40.58%, significantly lower than those of other varieties (lines). During 0-40 d of high-temperature treatment, fruit set per plant of MT and H36 were 12.00 and 7.57, while the fruit setting rates of H3 and MT at full bloom were 66.39% and 51.14% respectively, which were significantly higher than those of other varieties (lines). Correlation analysis showed that fruit setting rate was significantly positively correlated with SOD activity, POD activity, and pollen germination rate. Based on the comprehensive evaluation value of heat tolerance (D), the heat tolerance of the 12 passion fruit varieties (lines) was in the order of H36 > MT > Tainong No. 1 >H3 > P5 > P7 > Xiaji> P1 > Donghuimi > Yuanxiao > Huangyou No. 2 > DN. Cluster analysis results showed that the 12 varieties (lines) were divided into four groups, among which H36 and MT exhibited the strongest heat tolerance, while Huangyou No. 2 and DN showed the poorest. 【Conclusion】 Pollen germination rate, fruit setting rate, and fruits set number per plant can be used as comprehensive evaluation indicators for heat tolerance of different passion fruit varieties (lines). Two varieties (lines) with the strongest heat tolerance,H36 and MT, are screened.
