【Objective】 This study aimed to analyze the biological characteristics, active ingredient contents, and antioxidant activities of different monokaryotic strains of
Ganoderma sinense, to provide a theoretical basis for breeding excellent
Ganoderma sinense strains in Guangxi and the sustainable development of
Ganoderma industry.【Method】 Using the wild macrofungi from Guangxi as research objects, the strain SZ903 was obtained after multiple purification and identified via ITS sequence analysis. The optical microscope and fluorescence microscope were used to observe the monokaryotic, dikaryotic, and clamp connection of mycelia from single-colony spore germinations to identify monokar-yotic strains. The mycelium growth rate, density, pigment secretion, mycelial active components, and mycelial anti-oxidant activity of different monokaryotic strains and the strain SZ903 were investigated. Furthermore, correlation analyses were performed between mycelial active component content and antioxidant activity index.【Result】 The ITS sequences of strain SZ903 showed more than 99% similarity with 13
Ganoderma sinense strains, and the wild fungus was identified as
Ganoderma sinense. Only one strain had a single colony with clamp connections and adjacent dikaryons, indicating it was a dikaryotic strain. The nuclei of 30 strains of single colony mycelium were dispersed without clamp connection, indica-ting they were monokaryotic strains. Seven monokaryotic strains exhibited higher mycelial growth rates and greater density than strain SZ903, among which strain SZ903-8 had the fastest growth rate at 8.30 mm/d. The mycelial pigment secretion of strains SZ903-4 and SZ903-15 was higher. The total polysaccharide content of the seven monokaryotic strains was 1.07%-1.78%. The total polysaccharide contents of strains SZ903-6, SZ903-12, and SZ903-28 were 1.78%, 1.63%, and 1.57%, respectively, all higher than that of strain SZ903. The triterpenoid contents of strains SZ903-23 and SZ903-28 were higher than those of strain SZ903. The total flavonoid contents of strains SZ903-2, SZ903-8, and SZ903-23 were 1.02-1.25 mg/g, significantly higher than that of strain SZ903 (
P<0.05, the same below). The total antioxidant activity of strains SZ903-8 and SZ903-23 were 3.56 and 3.98 μmol/g, respectively, significantly higher than that of strain SZ903 (3.06 μmol/g). The SOD activities of seven monokaryotic strains were higher than those of strain SZ903. The hydroxyl radical scavenging rates of strains SZ903-2, SZ903-8, and SZ903-23 were significantly higher than that of strain SZ903 (47.1%). Correlation analysis results showed a highly significant negative correlation between total polysaccharide content and total flavonoid content (
P<0.01), with a correlation coefficient of -0.922, and a significant positive correlation between the total flavonoid content and the hydroxyl radical scavenging rate, with a correlation coefficient of 0.709.【Conclusion】 The wild macrofungi strain SZ903 from Guangxi is identified as
Ganoderma sinense. Strains SZ903-28 and SZ903-23 are screened from the seven
Ganoderma sinense monokaryotic strains with better mycelial growth indexes. The active component content and antioxidant activity of these two strains are higher, making them suitable for germplasm innovation and variety breeding of
Ganoderma sinense.
