MS-222对四指马鲅幼鱼麻醉效果、组织结构和肝脏抗氧化的影响

Effects of MS-222 on anesthetic effect, tissue structure and liver antioxidation of juvenile Eleutheronema tetradactylum

  • 摘要: 【目的】 研究MS-222对四指马鲅幼鱼麻醉效果、鳃组织损伤和肝脏抗氧化能力的影响,为四指马鲅抗逆性研究及MS-222作为水产品麻醉剂的使用提供科学参考。【方法】 以平均体长6.0±0.3 cm、平均体质量1.6±0.3 g的四指马鲅幼鱼为试验材料,以不添加麻醉剂、不充氧为对照,分析在30和35℃条件下不同质量浓度(10、15、20、25、30、40、50 mg/L)MS-222对四指马鲅幼鱼的静水麻醉效应,并根据其在MS-222麻醉溶液中的行为表现,进一步细化MS-222质量浓度;同时测定四指马鲅幼鱼肝脏的抗氧化酶活性,对其鳃组织进行观察。【结果】 在30和35℃条件下,随着MS-222质量浓度的增加,四指马鲅幼鱼进入同一麻醉时期的时间明显缩短;MS-222对幼鱼的最适麻醉质量浓度均为14 mg/L,且使用该浓度进行持续麻醉12 h后和复苏后24 h幼鱼存活率均为100.0%;MS-222不同浓度组肝脏抗氧化酶活性差异明显,其中过氧化氢酶(CAT)活性较对照升高,超氧化物歧化酶(SOD)活性较对照降低;观察幼鱼组织病理切片发现,随着MS-222质量浓度的增加,鳃小片会出现弯曲、末端肿大、严重时会断裂等现象,使用14 mg/L MS-222对幼鱼进行静水模拟运输第6 h时鳃小片会出现轻微的弯曲、水肿和排列紧密现象,但在模拟运输12 h后这些情况均得以改善。【结论】 不同质量浓度的MS-222均可对四指马鲅幼鱼产生镇静作用,30和35℃条件下四指马鲅幼鱼的适宜麻醉剂量均为14 mg/L,可降低鱼体对外界环境的应激反应。

     

    Abstract: 【Objective】 To investigate the effects of MS-222 on the anesthesia effects, gill tissue damage, and liver antioxidant capacity of juvenile Eleutheronema tetradactylum, which could provide scientific reference for research on stress resistance of Eleutheronema tetradactylum and the application of MS-222 as an aquatic product anesthetic. 【Method】 Using juvenile Eleutheronema tetradactylum with an average body length of 6.0±0.3 cm and an average body weight of 1.6±0.3 g as experimental material, with a control group receiving no anesthetic and no aeration, the staticwater anesthesia effects of different MS-222 mass concentrations(10, 15, 20, 25, 30, 40, 50 mg/L) on the juvenile Eleutheronema tetradactylum at 30 and 35 ℃ were analyzed. Based on their behavioral responses in the MS-222 anesthetic solution, the MS-222 mass concentration were further refined. Additionally, the antioxidant enzyme activity in the liver of juvenile Eleutheronema tetradactylum were measured and histological observations of their gill tissues were conducted. 【Result】 Under the conditions of 30 and 35 ℃, with the increase of MS-222 mass concentration, the time for juvenile Eleutheronema tetradactylum to enter the same anesthesia period greatly decreased; the optimal anesthesia mass concentration for juvenile Eleutheronema tetradactylum using MS-222 was 14 mg/L, and the survival rates of juvenile Eleutheronema tetradactylum after continuous anesthesia for 12 h and 24 h after recovery were 100.0%; the activities of liver antioxidant enzymes in different concentration groups of MS-222 showed obvious differences. Compared with the control, catalase(CAT) activity was increased, while superoxide dismutase(SOD) activity was decreased. Observing the pathological sections of juvenile Eleutheronema tetradactylum tissues, it was found that as the mass concentration of MS-222 increased, gill fragments exhibited bending, swelling at the end, and in severe cases, swelling and detachment. When using MS-222 at a concentration of 14 mg/L to simulate transportation in still water for 6 h, gill fragments showed slight bending, oedema, and tight arrangement. However, these conditions improved after 12 h of simulated transportation. 【Conclusion】 Different mass concentrations of MS-222 exhibit sedative effects on juvenile Eleutheronema tetradactylum. Under both 30 and 35 °C conditions, the optimal anesthetic dosage for juvenile Eleutheronema tetradactylum is determined to be 14 mg/L, which effectively reduces stress responses to external stimuli.
