Abstract:
【Objective】 The TCP gene family of
Xanthoceras sorbifolium was identified and its expression under salt stress was analyzed,its response mechanism to salt stress was studied,which provided theoretical basis for breeding of salt-tolerant germplasm of
X.sorbifolium.【Method】The TCP gene family of
X.sorbifolium was identified by bioinformatics method,and the protein physicochemical properties,phylogenetic evolution,conserved motifs,gene structure,chromo‐some localization and promoter cis-acting elements were analyzed.Real-time fluorescence quantitative PCR was used to detect the expression of TCP genes of
X.sorbifolium(
XsTCPs) in leaf and root under salt stress. 【Result】 A total of 24members of the TCP gene family(
XsTCP1-
XsTCP24) of
X.sorbifolium were identified,encoding 185-576 amino acids residues,protein molecular weight being 20576.21-63366.92 kD,theoretical isoelectric point being 4.62-10.25,all of which were hydrophilic proteins and located in the nucleus.Based on the phylogenetic analysis results,the 24
XsTCPs proteins could be divided into 3 subfamilies,among which the PCF subfamily contained 14 members,the CIN subfamily contained 6 members,and the CYC/TB1 subfamily contained 4 members.Except that
XsTCP23 and
XsTCP24 genes were unable to locate on the chromosome,the remaining TCP gene family members were unevenly distributed on 11 chromo‐somes in the form of single genes or gene clusters.A total of 10 conserved motifs were found in 24
XsTCPs proteins,among which Motif 1 was common to all proteins.The TCP gene family members of
X.sorbifolium contained light response elements,hormone response elements,stress response elements and plant growth and development elements.
XsTCP4,
XsTCP6,
XsTCP7,
XsTCP10,
XsTCP12,
XsTCP13,
XsTCP21 and
XsTCP22 genes contained drought-induced MYB binding elements.The results of real-time fluorescence quantitative PCR showed that the expression of the above genes changed greatly in the leaves and roots of
X.sorbifolium under salt stress,suggesting that the above 8
XsTCPs genes were involved in the process of salt stress response. 【Conclusion】 XsTCPs proteins may be involved in biological pro‐cesses by regulating various target genes involved in growth,development,metabolism and stress response.It is specu‐lated that 8
XsTCPs genes containing drought-induced MYB binding elements may help improve the resistance to salt stress.
