叶面喷施褪黑素对NaCl胁迫下南瓜幼苗生长和生理的调控作用

Effects of melatonin foliar spray on growth and physiology regulation of pumpkin seedlings under NaCl stress

  • 摘要: 【目的】探究不同浓度褪黑素对氯化钠(NaCl)胁迫下南瓜幼苗生长和生理特性的影响,为利用褪黑素提高南瓜幼苗的耐盐性提供理论依据。【方法】以南瓜杂交种银板栗为试验材料,采用盆栽试验,待南瓜幼苗长至3叶1心时以浇灌浓度为150 mmol/L NaCl模拟盐胁迫环境,设5个褪黑素喷施浓度处理,分别为10.0 μmol/L(T1)、20.0 μmol/L(T2)、50.0 μmol/L(T3)、100.0 μmol/L(T4)和150.0 μmol/L(T5);另设2个对照,分别为NaCl胁迫后不喷施褪黑素处理(CK)和非NaCl胁迫的清水浇灌处理(CK0)。盐胁迫当天于夜间无光照时间段开始喷施褪黑素溶液,之后每隔1 d喷施1次,分别于处理后0、6、12和18 d测定南瓜幼苗生长指标,处理后18 d测定光合指标、叶绿素含量、根长、根系活力及抗氧化系统指标。【结果】NaCl胁迫影响南瓜幼苗植株正常生长发育,经褪黑素处理后幼苗生长效果有不同程度的提升,其中,处理后18 d,T3处理南瓜幼苗的株高、茎粗、总叶绿素含量、根系活力、过氧化氢酶(CAT)活性提升效果最佳,分别较CK显著增加41.71%、41.08%、83.39%、46.77%和160.00%(P<0.05),鲜重、干重、干鲜比、超氧化物歧化酶(SOD)活性也有所提高,丙二醛(MDA)含量有所降低;与CK相比,低浓度褪黑素处理(T1和T2)对光合指标有改善作用,而高浓度褪黑素处理(T3、T4和T5)的南瓜叶片光合速率(Pn)低于CK。主成分分析结果表明,不同处理的主成分综合得分排名依次为CK0>T3>T4>T2>T1>T5>CK。【结论】叶面喷施适宜浓度的褪黑素可有效缓解NaCl胁迫对南瓜幼苗的损害,提高南瓜幼苗的耐盐性。综合南瓜幼苗生长和生理指标表现,以喷施50 μmol/L褪黑素对缓解南瓜幼苗NaCl胁迫伤害的作用较佳。

     

    Abstract: 【Objective】To explore the effects of different concentrations of melatonin on the growth and physiology characteristics of pumpkin seedlings under sodium chloride (NaCl) stress,which could provide theoretical basis for using melatonin to improve the salt tolerance of pumpkin seedlings.【Method】The pumpkin hybrid variety Silver Chestnut was used as the experimental material,and pot experiment was carried out.When the pumpkin seedlings grew to 3 leaves and1 bud,the salt stress environment was simulated by watering with 150 mmol/L NaCl.Five melatonin spraying concentrations were set as 10.0 μmol/L (T1),20.0 μmol/L (T2),50.0 μmol/L (T3),100.0 μmol/L (T4) and 150.0 μmol/L (T5),another 2 groups were set as control,no melatonin treatment after NaCl stress (CK) and water irrigation treatment without NaCl stress (CK0).On the day of salt stress,melatonin solution was sprayed at night during the period of no light,and then sprayed once every 1 d.The growth indexes of pumpkin seedlings were measured on 0,6,12 and 18 d after treatment,the photosynthetic indexes,chlorophyll content,root length,root activity and antioxidant system indexes were measured 18 d after treatment.【Result】NaCl stress affected the normal growth and development of pumpkin seedlings.After melatonin treatment,the growth effect of pumpkin seedlings was improved to varying degrees.Among them,18 d after treatment,T3 treated pumpkin seedlings showed the best improvement in plant height,stem diameter,total chlorophyll content,root activity and catalase (CAT) activity.Compared with CK,they were significantly increased by 41.71%,41.08%,83.39%,46.77%and 160.00%respectively (P<0.05);fresh weight,dry weight,dry-fresh ratio,superoxide dismutase (SOD) activity were also increased,and malondialdehyde (MDA) content was decreased.Compared with CK,low concentration melatonin treatments (T1 and T2) had an improved effect on photosynthetic indexes,while high concentration melatonin treatments (T3,T4 and T5) had lower photosynthetic rate (Pn) than CK.The results of principal component analysis showed that the ranking of principal component scores of different treatments was CK0>T3>T4>T2>T1>T5>CK.【Conclusion】Foliar spray of melatonin with appropriate concentration can effectively alleviate the damage of NaCl stress on pumpkin seedlings and improve the salt tolerance of pumpkin seedlings.Based on the growth and physiology indexes of pumpkin seedlings,spraying 50 μmol/L melatonin has the best effects on alleviating the injury of pumpkin seedlings under NaCl stress.
