Abstract:
【Objective】The objective of this study was to assess the effects of four fermented mixed residues(Siraitia grosvenorii residue, tofu residue, sweet potato peel and cocoyam powder) replacing part of diet on intestinal microflora of Fufeng Ma-chickens in Guangxi. At the same time, the optimal ratio of fermented mixed residue replacing part of diet was evaluated. and the reference basis for the application of four fermented mixed residue in chicken breeding was provided. 【Method】Twenty 60-day-old Fufeng Ma-chickens in Guangxi(female) were selected and randomly divided into 4 groups with 5 chickens in each group. The diets of 5%(R5),10%(R10),and 15%(R15) were replaced with fermented mixed residue of S. grosvenorii residue, tofu residue, sweet potato peel and cocoyam powder respectively. The whole diet was used as the control group(CK) in a feeding trial for 97 d. After feeding, the contents of the Fufeng Ma-chickens' large intestine were collected,and effects of fermented mixed residue replacing part of diet on the diversity index of intestinal microflora of Fufeng Ma-chickens in Guangxi were studied based on Illumina high-throughput sequencing technology.【Result】The Chao1 index of intestinal microflora of Fufeng Ma-chickens in Guangxi was ranked as R15>CK>R10>R5,Shannon index was ranked as CK>R15>R10>R5,and Simpson index was ranked as CK=R15=R10>R5. There was no significant difference in the Chao 1 index, Shannon index, and Simpson index between each experimental group and CK(
P>0.05, the same as below). At the level of phylum classification, among which Bacteroidetes,Firmicutes,and Proteobacteria were the dominant bacteria. The relative abundance of Bacteroidetes was ranked as R10>R15>CK>R5, Firmicutes was ranked as R5>CK>R15>R10, and Proteobacteria was ranked as CK>R10>R15>R5. Among them, the relative abundance of Proteobacteria and Epsilonbacteraeota in the R5 was significantly lower than that of CK(
P<0.05, the same as below). At the level of genus classification, the relative abundance of Proteus was rank as CK>R10>R5>R15.The relative abundance of the three alternative test groups was significantly lower than that of CK. The relative abundance of Ruminiclostridium_9 was ranked as R10>R15>CK>R5, and only the relative abundance of R10 group was significantly higher than that of the CK. The relative abundance of Faecalibacterium was ranked as CK>R10>R5>R15, and the relative abundance of R15 group was significantly lower than that of the CK. 【Conclusion】Replacing 5%, 10% and 15%diet by four fermented mixed residue of S. grosvenorii, tofu, sweet potato peel and cocoyam powder for Fufeng Machickens in Guangxi do not affect the species abundance and diversity of intestinal microflora of chickens. Based on the comprehensive evaluation of beneficial bacteria and harmful bacteria, 10% addition rate can increase the abundance of probiotic Ruminiclostridium_9 and has the least effect on Proteus, which is the optimal addition rate.
