湖羊m6A甲基转移酶METTL14基因CDS序列克隆及表达谱分析

Cloning and expression profile analysis of CDS sequence of m6A methyltransferase METTL14 gene from Hu sheep

  • 摘要: 【目的】克隆湖羊甲基转移酶样蛋白14基因(METTL14)编码区(CDS)序列,探究其分子特征及组织表达分布规律,为揭示METTL14基因调控湖羊肌肉发育的特异性分子机制打下基础。【方法】提取湖羊肌肉组织RNA并反转录合成cDNA,以cDNA为模板扩增METTL14基因CDS序列,通过ProtParam、ProtScale、SignaIP-6.0等在线软件进行生物信息学分析,并利用实时荧光定量PCR进行湖羊METTL14基因组织表达谱分析。【结果】湖羊METTL14基因CDS序列全长1371 bp,共编码456个氨基酸残基;湖羊METTL14基因CDS序列与黄牛、人类、小鼠、大鼠、猕猴、山羊、弯角剑羚、马、双峰驼、野牦牛、野猪和水牛等12个参考物种的METTL14基因核苷酸序列相似性为89.28%~99.78%,对应的METTL14氨基酸序列相似性为98.66%~100.00%;基于METTL14基因核苷酸序列相似性构建的系统发育进化树显示,湖羊与山羊的亲缘关系最近,与双峰驼的亲缘关系较远。湖羊METTL14蛋白分子式为C2277H3575N669O709S16,分子量为52179.45 Da,理论等电点(pI)为5.89,不存在信号肽,无跨膜结构域,具有较强的亲水性,属于不稳定蛋白,主要分布在细胞核(占69.6%);湖羊METTL14蛋白包含12个α-螺旋和10个β-折叠,但以无规则卷曲为主。METTL14基因在湖羊的心脏、肝脏、脾脏、肺脏、肾脏、大脑、睾丸、肌肉及皮下脂肪中均有表达,以心脏中的相对表达量最高,而肝脏中的相对表达量最低。【结论】METTL14作为m6A甲基化的核心蛋白,在物种进化过程中高度保守;METTL14基因在湖羊肌肉和脂肪等多个组织中均有表达,且表达量与机体的代谢活动相关,可作为调控湖羊肌肉生长发育的候选基因进一步研究。

     

    Abstract: 【Objective】Coding region (CDS) sequence of methyltransferase-like protein 14 gene (METTL14) in Hu sheep was cloned,which helped to explore its molecular characteristics and tissue expression distribution,and lay a foun-dation for revealing the specific molecular mechanism of METTL14 gene regulating muscle development in Hu sheep.【Method】RNA was extracted from the muscle tissue of Hu sheep and cDNA was synthesized by reverse transcription.The CDS sequence of METTL14 gene was amplified by using cDNA as template.Bioinformatics analysis was carried out by online softwares,such as ProtParam,ProtScale,and SignaIP-6.0.The tissue expression profile of METTL14 gene in Hu sheep was analyzed by real-time fluorescence quantitative PCR.【Result】The total length of the CDS region of Hu sheep METTL14 gene was 1371 bp,which encoded 456 amino acids residues.The CDS sequence of METTL14 gene in Hu sheep was 89.28%-99.78%similar to that in 12 reference species,including Bos taurus,Homo sapiens,Mus musculus,Rattus norvegicus,Macaca nemestrina,Capra hircus,Oryx dammah,Equus caballus,Camelus bactrianus,Bos mutus,Sus scrofa,Bubalus bubalis,and the corresponding amino acid sequence similarity of METTL14 was 98.66%-100.00%.The phylogenetic tree constructed based on the nucleotide sequence similarity of METTL14 gene showed that Hu sheep had the closest relationship with goats and far relationship with bactrian camels.The molecular formula of Hu sheep METTL14 protein was C2277H3575N669O709S16,the molecular weight was 52179.45 Da,and the theoretical isoelectric point(pI) was 5.89.There was no signal peptide and no transmembrane domain.It had strong hydrophilicity and was an un-stable protein,mainly distributed in the nucleus (69.6%).The METTL14 protein contained 12 α-helices and 10 β-folds,but it was mainly composed of random coils.The METTL14 gene was expressed in the heart,liver,spleen,lung,kid-ney,brain,testis,muscle,and subcutaneous fat of Hu sheep,with the highest relative expression in the heart and the lowest relative expression in the liver.【Conclusion】As the core protein of m6 A methylation,METTL14 is highly con-served during species evolution.The METTL14 gene is expressed in several tissues including muscle and fat of Hu sheep,and the expression level is related to the metabolic activity of the organism,which can be investigated as a candidate gene to regulate the muscle growth and development of Hu sheep.
