不同自发气调包装袋对菜心薹茎木质化的影响

Effects of different spontaneous modified atmosphere packaging bags on lignification of bolting stem of Chinese flowering cabbage

  • 摘要: 【目的】探究自发气调包装和市购聚乙烯膜包装对菜心薹茎木质化的影响,并分析细胞壁相关酶基因的表达水平,以期为菜心保鲜贮运和木质化调控机理研究提供理论依据。【方法】以宁夏四九菜心为试验材料,分别测定常温(25℃)贮藏期间(8 d)3种包装袋自发气调包装(MP20、MP30)和聚乙烯薄膜(对照)包装菜心薹茎的生理指标,以及包装袋内的O2和CO2含量,并通过实时荧光定量PCR分析薹茎纤维素酶基因(BrCEL3)、木葡聚糖转葡糖基酶基因(BrXET2、BrXET3)和β-甘露聚糖酶基因(BrMAN1、BrMAN5和BrMAN6)的表达情况。【结果】贮藏期间MP20和MP30自发气调包装袋能有效延缓菜心薹茎硬度增加,减少水分流失;袋内气体成分变化,形成低O2高CO2的环境;使薹茎在贮藏结束时尚含有较高的可溶性蛋白含量(0.54和0.93 mg/g)和维生素C含量(0.45和0.62 mg/g),抑制木质素合成关键酶苯丙氨酸解氨酶(PAL)和过氧化物酶(POD)活性的增加;减少木质素含量积累和纤维素增加,至贮藏结束,MP20和MP30气调包装的薹茎木质素含量分别为0.28和0.27 mg/g,较对照分别降低12.50%和15.63%,而纤维素含量分别为4.16和4.60 mg/g,较初始值分别降低32.57%和25.41%;抑制丙二醛(MDA)含量增加,贮藏至4~8 d时MDA含量显著低于对照(P<0.05);BrCEL3BrMAN5、BrXET2BrXET3、BrMAN1BrMAN6基因相对表达量的变化趋势两两类似,且均低于对照(MP20的第4 d除外)。【结论】自发气调包装可有效减少常温贮藏期间菜心薹茎的木质素和纤维素含量积累,减缓木质化进程。经综合分析,MP30气调包装对菜心薹茎的贮运保鲜及减缓木质化更有利。

     

    Abstract: 【Objective】The effects of modified atmosphere packaging and commercial polyethylene film packaging on lignification in bolting stem of Chinese flowering cabbage and the expression patterns of cell wall related enzyme genes were investigated to provide a theoretical basis for the study of preservation,storage and lignification regulation mechanism of Chinese flowering cabbage.【Method】Ningxia Sijiu flowering cabbage was as the material.The physiological indexes and the contents of O2 and CO2 of Chinese flowering cabbage packaged in three kinds of packaging bagsmodified atmosphere packaging (MP20,MP30) and commercial polyethylene film packaging(control)were determined respectively,during storage period(8 d) at room temperature(25℃).And the expression pattern of cellulase gene(BrCEL3),xyloglucan transglucosylase (BrXET2 and BrXET3) and β-mannanase enzyme gene (BrMAN1,BrMAN5 and BrMAN6) were analyzed by real-time fluorescence quantitative PCR.【Result】During storage,MP20 and MP30 modified atmosphere packaging bags could effectively delay the increase of stem hardness and reduce water loss.And the composition of the gas in the bag changed,forming a low O2 and high CO2 atmosphere.At the end of storage,the bolting stems in MP20and MP30 groups were still contained higher soluble protein content (0.54 and 0.93 mg/g) and vitamin C content (0.45and 0.62 mg/g).Modified atmosphere packaging inhibited the increase of activities of phenylalanine ammonia-lyase(PAL) and peroxidase(POD),which were the key enzymes in lignin synthesis,and also reduced the accumulation of lignin and cellulose content.At the end of storage,the lignin contents of the bolting stems in MP20 and MP30 modified atmosphere packagings were 0.28 and 0.27 mg/g,decreased by 12.50%and 15.63%compared with the control,respectively,while the cellulose contents were 4.16 and 4.60 mg/g,decreased by 32.57%and 25.41%compared with the initial value,respectively.Modified atmosphere packaging inhibited the increase of malondialdehyde(MDA) content,the content of MDA was significantly lower than that of control group after 4 to 8 d of storage (P<0.05).The expression trends of BrCEL3 and BrMAN5,BrXET2 and BrXET3,BrMAN1 and BrMAN6 genes were similar,and the expression levels of these genes in modified atmosphere packaging were lower than control group (except for day 4 in MP20 group).【Conclusion】Modified atmosphere packaging effectively reduces the accumulation of lignin and cellulose in bolting stem of Chinese flowering cabbage,and slows down the lignification process.Through comprehensive analysis,the MP30 modified atmosphere packaging is more beneficial to the preservation and slowing down the lignification of bolting stems of Chinese flowering cabbage.
