黄花倒水莲内生真菌的分离鉴定及抑菌活性研究

Isolation, identification and antimicrobial activity of endophytic fungi from Polygala fallax Hemsl

  • 摘要: 【目的】从野生黄花倒水莲(Polygala fallax Hemsl)中分离纯化对植物病原菌具有拮抗作用的内生真菌并进行鉴定,为后续进一步发掘黄花倒水莲内生真菌潜在利用价值提供理论依据。【方法】采用组织块分离法从健康的野生黄花倒水莲中分离纯化内生真菌,结合形态学和分子生物学方法对其进行分类鉴定;采用平板对峙法筛选对三七根腐病菌(Fusarium solani)、叶点霉(Phyllosticta zingiberi)、苦玄参叶斑病菌(Scrophularia ningpocnsis)、水稻恶苗病菌(F. moniliforme Sheld.)、烟草灰霉病菌(Botrytis cinerea Pevs.)和黑曲霉(Aspergillus niger) 6种植物病原菌具有强抑菌作用的活性菌株。【结果】分别从17株野生黄花倒水莲的根、茎和叶中分离纯化得到99、183和109株内生真菌;经鉴定,共划分为62个分类单元,其中57个分类单元隶属于真菌界2门4纲12目22科33属,5个未确定分类地位。黄花倒水莲内生真菌总定殖率和分离率分别为77.17%和40.94%,其中茎部内生真菌的定殖率(36.44%)和分离率(19.16%)均明显高于根部(31.94%和10.37%)和叶片(8.80%和11.41%)。抑菌活性研究结果表明,共有8株内生真菌对4种以上供试植物病原菌的抑菌率在50.00%以上;其中,LCRPH-27对三七根腐病菌的抑制作用最强,抑制率为40.47%; LCSMBA-1对叶点霉和黑曲霉的抑制作用最强,抑制率分别为68.07%和65.37%; LCSMBA-153对苦玄参叶斑病菌的抑制作用最强,抑制率为75.60%; LCRPH-9对烟草灰霉病菌的抑制作用最强,抑制率为88.50%; LCLF-88对水稻恶苗病菌的抑制作用最强,抑制率为81.23%。【结论】黄花倒水莲含有丰富的内生真菌资源,具有良好而广谱的抑菌活性,且其代谢产物在生产天然药物或生物防治等方面有巨大的潜在开发应用价值。

     

    Abstract: 【Objective】Endophytic fungi with antagonistic effects were isolated,purified and identified from the wild Polygala fallax Hemsl,which provided theoretical basis for further exploring the potential utilization value of endophytic fungi of P. fallax.【Method】The endophytic fungi were isolated and purified from wild and healthy P. fallax by tissue block separation method,and were classified and identified by morphology and molecular biology. Plate confrontation method was used to screen and determine the activity of endophytic fungi against Fusarium solaniPhyllosticta zingiberiScrophularia ningpocnsisF. moniliforme Sheld., Botrytis cinerea Pevs. and Aspergillus niger.【Result】In this study,99, 183 and 109 strains of endophytic fungi were isolated and purified from the roots,stems and leaves of 17 wild P. fallax. After identification,it was divided into 62 taxa,among which 57 taxa belonged to 33 genera,22 families,12 orders,4 classes and 2 phyla,and the rest 5 were undetermined taxonomic status. The total colonization rate and isolation rate were 77.17% and 40.94% respectively. The colonization rate and isolation rate of endophytic fungi in stem(36.44% and 19.16%)were significantly higher than those in root(31.94% and 10.37%)and leaf(8.80% and 11.41%). The results showed that 8 strains of endophytic fungi had more than 50.00% inhibition rate to more than 4 kinds of plant pathogens. Among them,LCRPH-27 had the strongest inhibitory effect on F. solani,with the inhibition rate of 40.47%. LCSMBA-1 had the strongest inhibitory effect on P. zingiberi and A. niger with the inhibition rates of 68.07% and 65.37% respectively. LCSMBA-153 had the strongest inhibitory effect on S. ningpocnsis with the inhibition rate of 75.60%. LCRPH-9 had the strongest inhibitory effect on B. cinerea with the inhibition rate of 88.50%. LCLF-88 had the strongest inhibitory effect on F. moniliforme with the inhibition rate of 81.23%.【Conclusion】The endophytic fungi of P. fallax are rich in resources and diversity,and have good antibacterial activity. And its metabolites have great potential development and application value in the production of natural drugs or biological control.
