不同添加剂对凌云乌鸡生长性能、免疫功能及盲肠微生物多样性的影响

Effects of different additives on growth performance, immune function and microbial diversity in cecum of Lingyun black bone chicken

  • 摘要: 【目的】探究不同添加剂对凌云乌鸡生长性能、免疫器官指数、血清免疫指标及盲肠微生物多样性的影响,为凌云乌鸡饲养过程中选择替抗产品提供参考依据。【方法】选取1日龄凌云乌鸡500只,公母各半,随机分为5组[5个处理,分别为添加剂组的T1~T4处理和对照(CK)],饲喂基础饲粮,T1、T2和T4处理分别在试验鸡常规饮水中添加3.0%复合乳酸菌、3.5%枯草芽孢杆菌和0.5%复方中药,T3处理在基础饲粮中添加0.3 mg/kg有机硒(以硒计),测定分析1~6周龄试验鸡的生长性能及6周龄试验鸡的免疫器官指数、血清免疫指标和盲肠微生物多样性。【结果】 1周龄凌云乌鸡的平均日采食量,T2、T3、T4处理和CK均显著低于T1处理(P<0.05,下同),料重比以T1和T4处理显著低于T2、T3处理和CK; 2周龄凌云乌鸡的平均日增重,CK和T1处理显著高于T2、T3和T4处理,料重比以T2和T3处理显著低于T1、T4处理和CK; 3~6周龄凌云乌鸡的平均日增重、平均日采食量和料重比均无显著差异(P>0.05,下同)。T1~T4处理凌云乌鸡的脾脏指数较CK均有所提高,T1和T3处理的法氏囊指数均显著高于CK,T2和T4处理的法氏囊指数与CK差异不显著,T1和T3处理的胸腺指数较CK分别提高13.11%和14.75%。T1~T4处理凌云乌鸡的免疫球蛋白A和免疫球蛋白M较CK均有所提高,T1、T2和T4处理的免疫球蛋白G均较CK有所提高,且以T1处理最高。各处理凌云乌鸡间的Shannon指数、Simpson指数和Chao1指数差异不显著,但T1处理较CK及其他处理均有所提高,且T1处理的Pielou均匀度显著高于CK和T3处理。在门水平,T1、T2、T3、T4处理和CK凌云乌鸡盲肠细菌相对丰度>1.00%的优势菌门总数分别为5、6、5、7和5个,且优势菌门的总相对丰度分别为95.92%、98.11%、95.96%、97.97%和97.10%。在属水平,T1、T2、T3、T4处理和CK凌云乌鸡盲肠的细菌优势菌属总数分别为18、21、19、19和19个,且优势菌属的总相对丰度分别为80.46%、83.11%、81.58%、79.23%和81.31%。【结论】饲喂复合乳酸菌、有机硒、复方中药和枯草芽孢杆菌均可提高凌云乌鸡的平均日增重,降低其料重比,提高免疫器官指数和血清免疫指标,同时提高盲肠微生物多样性及群落相对丰度,尤其以饲喂复合乳酸菌的效果更佳。

     

    Abstract: 【Objective】 This experiment was conducted to investigate the effects of different additives on growth performance, immune organ indexes, serum immune indexes and microbial diversity in cecum of Lingyun black bone chicken to provide reference for the selection of antifungal products in Lingyun black bone chickens.【Method】 Five hundred oneday-old Lingyun black bone chicken were randomly divided into 5 groups[(5 treatments, T1-T4 added additives and control(CK)]. Chickens in the CK were fed a basal dietand normal water, and those in the T1, T2, T4were fed the water with 3.0% compound Lactobacillus, 3.5% Bacillus subtilis, organic selenium 0.5% Chinese herbal medicine, respectively. T3 were fed a basal diet with 0.3mg/kg organic selenium (as selenium). The growth performance of chickens aged from 1 to 6 weeks and immune organ index,serum immune index, cecum microbial diversity at 6 weeks were measured.【Result】 The results showed as follows: at 1week of age, the average daily feed intake (ADFI)of CK, T2, T3, T4 were significantly lower than those of T1(P<0.05, the same below), the feed conversion ratio(F/G)of T1 and T4 were significantly lower than those of T2, T3 and CK. At 2 weeks of age, the average daily gain (ADG)of C and T1 were significantly higher than others of T2, T3 and T4, the F/Gof T2 and T3 were significantly lower than those of CK,T1 and T4. At 3-6 weeks of age, there was no significant differences in the ADG(P>0.05, the same below), the ADFI, the feed conversion ratio between CK and treatment groups. Compared with CK, the spleen indexes of treatment groups increased, bursa of fabricius indexes of T1, T3 were significantly increased, bursa of fabricius indexes of T2, T4 had no significant differences compared with CK, thymus indexes of T1, T3 increased by 13.11% and 14.75% compared with CK respectively. Compared with CK, the immunoglobulin A, immunoglobulin M of treatment groups increased, the immunoglobulin G of T1, T2 and T4 increased, and the T1 was the highest. There was no significant differences in Shannon index, Simpson index, Chao1index among treatment groups, but those of T1 increased compared with CK and other treatments, Pielou’s evenness of T1 were significantly higher than that of CK and T3. At the phylum level, the number of the dominant bacteria whose relative abundance was over 1.00% of T1, T2, T3,T4 and CK were 5, 6, 5, 7 and 5,with relative abundance of 95.92%, 98.11%, 95.96%, 97.97% and 97.10%. At the genus level, the number of the dominant bacteria of T1, T2, T3, T4 and CK were 18, 21, 19, 19 and19, with relative abundance of 80.46%, 83.11%, 81.58%, 79.23%, 81.31%.【Conclusion】 It is concluded that, adding compound Lactobacillu, organic selenium, Chinese herbal medicine and B. subtilis in water can increase ADG decrease feed conversion ratio,improve immune organ indexes, serum immune indexes,and improve the cecalmicrobial diversity and relative abundance of microorganisms of Lingyun black bone chicken, and the compound Lactobacillus has the optimal effects.
