Abstract:
【Objective】Studying on the mechanism of the combination of resveratrol(RES)and epigallocatechin-3-gallate(EGCG)on the antioxidant capacity of skeletal muscle in mice,in order to develop a theoretical basis for the development of new,green and safe feed additive which could improve meat quality by enhancing the antioxidant capacity of skeletal muscle in livestock and poultry.【Method】Forty cleaning grade Kunming mice weighing 18-22 g were randomly divided into four groups,namely the dimethyl sulfoxide(DMSO)control group,the RES group,the EGCG group and the RES + EGCG combination group. Ten mice in each group were administered by gavage at a dose of 400 mg/kg per day,and the antioxidant enzyme activity indexes were measured in the skeletal muscle of mice after 28 d of continuous administration,and the expression of antioxidant-related genes in the skeletal muscle of mice was determined by fluorescence quantitative PCR.【Result】There was no significant effect on food intake,body weight and organ index in mice treated with RES in combination with EGCG(
P>0.05). Compared with DMSO,malondialdehyde(MDA)levels in serum and muscle were significantly lower in the RES,EGCG and RES+EGCG groups(
P<0.05,the same below),glutathione peroxidase(GSH-Px)and total superoxide dismutase(T-SOD)activities were significantly higher,GSH-Px,superoxide dismutase 1(SOD1),superoxide dismutase 2(SOD2)and nuclear factor NF-E2-related factor(
Nrf2)gene expression levels were significantly increased and Kelch-like ECH associated protein 1(Keap1)gene expression levels were significantly decreased in the gastrocnemius of mice. Compared with RES or EGCG alone,the serum levels of
GSH-Px and T-SOD were significantly higher,MDA levels were significantly lower,and GSH-Px gene expression levels were significantly higher in gastrocnemius muscle;the expression levels of SOD1 gene were significantly higher in the combined group compared with the EGCG group,and the expression levels of
SOD2 gene were significantly higher compared with that of RES group.【Conclusion】The combination of RES and EGCG significantly increases the activity of antioxidant enzymes and the expression of related antioxidant genes in mice,and may activate the Keap1-Nrf2 signaling pathway,indicating that the combination of RES and EGCG can improve the antioxidant level in muscles.
