Na+、K+、pH及3种单糖对湖拟鲤精子活力的综合调控

Comprehensive regulation of sperm motility of Rutilus rutilus lacustris by Na+,K+,pH,and 3 monosaccharides

  • 摘要: 【目的】明确Na+、K+、pH及3种单糖对湖拟鲤精子活力的单独调控和综合调控,筛选出湖拟鲤精子的适宜激活配方,为其人工繁殖及种质资源保护提供技术参考。【方法】以快速运动时间(FT)和寿命时间(LT)为指标,采用单因素试验和正交试验研究不同渗透压条件下Na+、K+、pH及3种单糖(葡萄糖、果糖和半乳糖)对湖拟鲤精子活力的单独和综合调控作用,筛选湖拟鲤精子的适宜激活液配方。【结果】 Na+溶液渗透压为282.36 kPa时湖拟鲤精子的FT和LT达最大值(40.00和285.33 s),K+溶液渗透压为211.77 kPa时湖拟鲤精子的FT和LT达最大值(36.00和130.33 s),葡萄糖溶液渗透压为352.95 kPa时湖拟鲤精子的FT和LT达最大值(43.67和355.33 s),果糖溶液渗透压为352.95 kPa时湖拟鲤精子的FT和LT达最大值(45.00和372.67 s),半乳糖溶液渗透压为235.30 kPa时湖拟鲤精子的FT和LT达最大值(40.00和357.00 s),pH为8.0时湖拟鲤精子的FT和LT达最大值(30.00和100.33 s)。正交试验结果表明,湖拟鲤精子活力综合调控的最优配方为果糖溶液渗透压117.65 kPa、pH 8.0、Na+溶液渗透压188.24 kPa,对应的FT和LT分别是58.33和370.00 s;果糖、pH和Na+对湖拟鲤精子活力的调控力度为果糖>Na+>pH,且表现为果糖和Na+对湖拟鲤精子的FT和LT调控效果有显著影响(P<0.05),而pH的影响不显著(P>0.05)。【结论】适宜浓度的Na+、K+、葡萄糖、果糖及半乳糖对湖拟鲤精子活力均具有较好的促进作用,主要是通过改变渗透压来调控湖拟鲤精子活力;适宜的pH为7.5~8.5。通过正交试验筛选出的湖拟鲤精子最优激活配方为果糖溶液渗透压117.65 kPa、pH 8.0、Na+溶液渗透压188.24kPa,其调控效果优于单因素最优渗透压,调控力度为果糖>Na+>pH。

     

    Abstract: 【Objective】 To clarify the individual and comprehensive regulation of sperm motility of Rutilus rutilus lacustris by Na+,K+,pH and 3 monosaccharides,to screen the suitable formula of the sperm activating liquid of R.rutilus lacustris,so as to provide technical reference for artificial reproduction and genetic resource conservation of the R.rutilus lacustris.【Method】 Fast movement time(FT)and life time(LT)were taken as indicators,and single factor test and orthogonal test were used to study individual and comprehensive regulation effects of Na+,K+,pH and 3 monosaccharides(glucose,fructose and galactose)on sperm motility of the R.rutilus lacustris under different osmotic pressure conditions to form a suitable formula of the sperm activating liquid of R.rutilus lacustris.【Result】 When Na+ solution osmotic pressure was 282.36 kPa,the FT and LT of R.rutilus lacustris sperm reached the maximum(40.00 and 285.33 s).When K+ solution osmotic pressure was 211.77 kPa,the FT and LT of R.rutilus lacustris sperm reached the maximum(36.00 and 130.33 s).When glucose solution osmotic pressure was 352.95 kPa,the FT and LT of R.rutilus lacustris sperm reached the maximum(43.67 and 355.33 s).When fructose solution osmotic pressure was 352.95 kPa,the FT and LT of R.rutilus lacustris sperm reached the maximum(45.00 and 372.67 s).When galactose solution osmotic pressure was 235.30 kPa,the FT and LT of R.rutilus lacustris sperm reached the maximum(40.00 and 357.00 s).When pH was 8.0,the FT and LT of R.rutilus lacustris sperm reached the maximum(30.00 and 100.33 s).Orthogonal test results showed that the optimal formula for comprehensive regulation of sperm motility of the R.rutilus lacustris was: Fructose solution osmotic pressure of 117.65 kPa,pH of 8.0,Na+ solution osmotic pressure of 188.24 kPa,FT of 58.33 s and LT of 370.00 s.The regulation intensity was fructose>Na+>pH.The results showed that fructose and Na+had significant effects on FT and LT regulation of R.rutilus lacustris sperm(P<0.05),but the effect of pH was not significant(P>0.05).【Conclusion】 Appropriate concentrations of Na+,K+,glucose,fructose and galactose promotes sperm motility of the R.rutilus lacustris mainly through changing osmotic pressure.The suitable pH is 7.5-8.5.Through orthogonal test,the optimal activating formula of the R.rutilus lacustris sperm is: Fructose of 117.65 kPa,pH of 8.0,and Na+ of 188.24 kPa,and regulation effect of this formula is better than that of the single-factor optimal osmotic pressure.The regulation intensity is: fructose>Na+>pH.
