芒果不同砧穗组合早期嫁接亲和性综合评价
Comprehensive evaluation of grafting compatibility of different stock-scion combinations in mango

摘要: 【目的】通过测定芒果不同砧穗组合嫁接苗生长指标及砧穗间生理生化相似程度等指标,筛选早期亲和的砧穗组合,为芒果优良品种配套砧木的选择提供参考依据。【方法】以金煌芒(J)、台农1号(T)、凯特芒(K)、贵妃芒(G)和热农1号(R)等5个芒果品种为接穗,紫花芒3号(Z)、红象牙芒(X)、广东土芒(G)、海南土芒(H)和鹰嘴芒(Y)为砧木,设置25个砧穗组合,测定各组合嫁接苗生长指标及砧穗间生理生化相似程度,通过主成分分析、隶属函数法和聚类分析对各组合的嫁接亲和性进行综合评价及聚类。【结果】各砧穗组合间成活率以ZG组合最低(31.85%),其他组合的成活率均高于50.00%;GK和YT组合的穗砧粗度比为1.00,ZG组合的穗砧粗度比(0.83)与1相差最大;各组合嫁接苗生长指标显示ZT、GT和YT组合嫁接苗生长旺盛,ZG和HG组合生长差;部分组合间生理生化相似程度存在显著差异(P<0.05),其中,可溶性蛋白和可溶性糖含量相差最小的组合分别为HK和ZT,砧穗叶片多酚氧化酶(PPO)活性及过氧化物酶(POD)活性相差最小的组合分别是ZK和YT。主成分分析从10个指标中筛选出4个主要相关指标,分别为新梢长、成活率、砧穗|可溶性糖含量差|和砧穗|POD活性差|;隶属函数法分析得出各主栽品种嫁接组合的亲和性强弱顺序,分别为:金煌芒GJ>YJ>HJ>XJ>ZJ;台农1号YT>GT>ZT>XT>HT;凯特芒GK>ZK>XK>YK>HK;贵妃芒GG>YG>XG>HG>ZG;热农1号GR>XR>HR>ZR>YR。聚类分析将砧穗组合分为4类,嫁接亲和性强有1个,为鹰嘴芒/台农1号组合(YT),亲和性较强有12个组合,亲和性中等有11个组合,不亲和1个,为紫花芒3号/贵妃芒组合(ZG)。【结论】新梢长、成活率、砧穗|可溶性糖含量差|和砧穗|POD活性差|可作为芒果砧穗组合嫁接亲和性的评价指标。金煌芒宜采用鹰嘴芒和广东土芒作砧木,台农1号宜采用鹰嘴芒作砧木,凯特芒、热农1号和贵妃芒宜采用广东土芒作砧木。贵妃芒不宜采用紫花芒3号作砧木。Abstract: 【Objective】To determine the growth indicators of stock-scion combinations in mango grafted seedlings and physio-biochemical similarity between rootstocks and to screen stock-scion combinations with good early compatibility,so as to provide references for the rootstock selection of improved mango varieties.【Method】Five mango varieties,Jinghuang(J),Tiannong No.1(T),Kaite(K),Guifei(G)and Renong No.1(R)were used as scions and Zhihua No.3(Z), Hongxiangya(X),Guangdongtumang(G),Hainantumang(H)and Yingzuimang(Y)were taken as rootstocks to make 25 stock-scion combinations. Some growth indexes of grafted seedlings and physio-biochemical similarity of rootstocks-scions were determined. And grafting compatibility of stock-scion combinations were evaluated through principal component analysis,membership function method and cluster analysis.【Result】ZG combination had the worst survival rate(31.85%), while the survival rate of other combinations were all above 50.00%. Scion/rootstock diameter ratios of GK and YT combinations were 1.00,that of ZG combination was 0.83,which was the maximum difference from 1. The growth indexes of combinations indicated that grafted seedling of ZT,GT and YT combinations grew vigorously,while ZG and HG combinations had poor growth potential. There were significant differences in physio-biochemical similarity among some combinations(P<0.05). The least absolute value of soluble protein content difference and soluble sugar content difference was observed in HK and ZT combination,the ZK and YT combinations showed the absolute value of polyphenol oxidase(PPO) and peroxidase(POD)activity difference. Four main related indexes were screened from the 10 indexes by principal component analysis,which were new shoot length,survival rate,the absolute value of soluble sugar content difference and POD activity difference between rootstock and scion. The result of membership function method showed the order of the graft compatibility of combinations. Jinghuang stock-scion combinations from high to low was GJ>YJ>HJ>XJ>ZJ, Tainong No.1 was YT>GT>ZT>XT>HT,Kaite was GK>ZK>XK>YK>HK,Guifei was GG>YG>XG>HG>ZG,and Renong No.1 was GR>XR>HR>ZR>YR. The cluster analysis showed that 25 stock-scion combinations were divided into 4 clusters,including one combination,namely YT combination,in group high compatibility,and twelve combinations in group relatively high compatibility,eleven combinations in group medium compatibility and one combination,namely ZG combination,in group incompatibility.【Conclusion】The new shoot length,survival rate,the absolute value of soluble sugar content difference and POD activity difference between rootstocks and scions are credible indexes for evaluation of early graft compatibility in mango. It is suggested that Tainong No.1 can be grafted with Yingzui;Jinghuang can be with Yingzui and Guangdongtumang,Kaite,Renong No.1 and Guifei can be with Guangdongtumang. Guifei can not be with Zihua No.3.
