桃果实ζ-胡萝卜素脱氢酶基因克隆及表达分析

Cloning and expression analysis of ζ-carotene desaturase gene in peach fruit

  • 摘要: 目的克隆桃果实ζ-胡萝卜素脱氢酶基因(PpZDS)的全长序列,并对其进行生物信息学及表达分析,为研究桃果实类胡萝卜素生物合成的分子机理提供参考.方法从黄肉品种桃果实中克隆PpZDS基因,对其进行生物信息学分析,并通过实时荧光定量PCR(qRT-PCR)检测其在桃果实不同成熟期的表达情况.结果克隆获得的PpZDS基因全长2014 bp,具有完整的编码区,长度为1716 bp,编码571个氨基酸.PpZDS蛋白含有ZDS特征序列(KVAIIGA-GLAGMSTAVELLDQGHEVDIYESR),属于NAD_binding_8超级家族保守结构域.PpZDS蛋白与同为蔷薇目的苹果(gi|33313474)和草莓(gi|256041892)的ZDS蛋白亲缘关系较近,物种间分化程度较小.整个桃果实成熟期PpZDS基因在果肉和果皮中均有表达,从软核期至硬熟期均呈逐渐升高趋势,硬熟期达最高,完熟期降低;不同桃果实发育期果肉中的PpZDS基因表达量均大于果皮中表达量,其中硬核期、硬熟期和完熟期的桃果实中PpZDS基因在果肉中的表达量显著高于果皮中的表达量(P<0.05).结论PpZDS基因表达对桃果实类胡萝卜素生物合成及呈色发挥正向调控作用.

     

    Abstract: ObjectiveFull-length sequence ofζ-carotene desaturase(PpZDS)gene in peach fruit was cloned and its bioinformatics analysis and expression analysis were also conducted to provide reference for the study of biosynthetic mo-lecular mechanism of carotene organisms in peach fruits.MethodPpZDS gene in peach fruit from yellow peach variety was tested and its bioinformatics analysis was carried out. Real-time fluorescence quantitative PCR(qRT-PCR)was used to test its expression in different developmental stages.ResultThe cloned gene PpZDS was 2014 bp in full length,it con-tained complete coding region of 1716 bp,and encoded 571 amino acids. PpZDS protein contained the characteristic se-quence of ZDS(KVAIIGAGLAGMSTAVELLDQGHEVDIYESR),which belonged to the NAD_binding_8 super family-conserved domains. PpZDS protein showed close genetic relationship with ZDS protein in apple(gi|33313474)and straw-berry(gi|256041892)which belonged to Rosales order and differentiation degree between different species was low. Gene PpZDS showed expression in both flesh and peel during whole peach fruit maturation period,and the expression showed a tendency of gradual increase from soft-core stage to firm-ripe stage,reaching the highest at firm-ripe stage,and then de-creased at full ripe stage. The expression of gene PpZDS in the fresh was generally higher than that in the peel at different maturation periods. When peach fruit was at firm-core stage,firm-ripe stage and full ripe stage,expression of gene PpZDS in the fruit fresh was significantly higher than the expression in the peel(P<0.05).ConclusionThe expression of gene PpZDS can be involved in the positive regulation of peach fruit carotenoid biosynthesis and color appearance.
