广西不同产地石韦药材绿原酸含量的测定
Determination of chlorogenic acid content in Pyrrosia produced from different areas of Guangxi

摘要: 【目的】采用高效液相色谱法(HPLC)测定分析广西不同产地石韦药材绿原酸含量的差异,为广西石韦药材资源的合理开发与利用提供参考。【方法】采用HPLC测定广西不同产地石韦药材绿原酸的含量,具体的色谱条件:色谱柱为C18,流动相为乙腈-0.05%磷酸(11.5∶88.5),检测波长326 nm,流速1.0 mL/min,柱温30℃。【结果】绿原酸在0.0368~0.8096μg范围内线性关系良好,回归方程为y=2607.6x+57.476(R2=0.9999),平均回收率为100.08%,RSD=0.72%。广西不同产地石韦药材绿原酸含量存在较大差异,其中,以采自金秀县罗香乡的石韦绿原酸含量最高,为3.6967 mg/g;其次是采自资源县宝鼎瀑布景区和资源县龙洞峡景区的石韦,其绿原酸含量分别为3.4678和3.1676 mg/g;绿原酸含量最低的是靖西县柔软石韦(1.1326 mg/g)。【结论】以HPLC测定石韦绿原酸具有操作简便、结果准确、重现性好等优点,是鉴别石韦品种及评价石韦药材质量的重要检测方法。广西不同产地石韦药材绿原酸含量存在较大差异,可能与产地、品种及采收时间等因素有关。Abstract: ObjectiveUsing HPLC method, the content of chlorogenic acid in Pyrrosia produced from different areas of Guangxi was determined to provide references for appropriate utilization and development of Pyrrosia medicinal materials in Guangxi. MethodThe content of chlorogenic acid in Pyrrosia was determined by HPLC. Chromatographic conditions were as follows. Chromatographic column was C18. Mobile phase was acetonitrile-0.05% phosphoric acid (11.5∶88.5). Detection wavelength was 326 nm. Flow rate was 1.0 mL/min. Column temperature was 30℃. ResultThere was good linear relation-ship of chlorogenic acid in the range of 0.0368-0.8096μg. The regression equation was y=2607.6x+57.476 (R2=0.9999). Av-erage recovery rate was 100 . 08% ( RSD=0 . 72%) . Vast differences of chlorogenic acid were observed in Pyrrosia from different areas of Guangxi. The highest content of chlorogenic acid was found in Pyrrosia from Luoxiang Village of Jinxiu County (3.6967 mg/g) followed by Pyrrosia from Baoding Waterfall and Dragon Cave scenic region of Ziyuan County with the chlorogenic acid content of 3.4678 and 3.1676 mg/g, respectively. The lowest content of chlorogenic acid was shown in Pyrrosia porosa from Jingxi County (1.1326 mg/g).ConclusionThe HPLC method of measuring chlorogenic acid was simple, accurate and reproductive, thus it’s an important detection method for identifying and evaluating Pyrrosia medicinal materials. The con-tent of chlorogenic acid varied in Pyrrosia from different areas of Guangxi, which may be caused by origin, cultivar or harvest time.
